摘  要:研究冷冻保存(-196℃)对野牦牛精子顶体酶等酶活性的影响。应用Giemsa染色法、BAPNA底物法、改良明胶底膜法和磷酸苯二钠法分别测定不同保存年份的精子顶体完整率、顶体酶、透明质酸酶和酸性磷酸酶活性;采用酶动力学分光光度法,分别以丙酮酸钠和α-已酮酸钠为底物测定乳酸脱氢酶及其同工酶C4活性,并计算C4的相对活性。结果表明,在冷冻保存条件下,野牦牛精子有关酶活性下降;并随保存时间的延长,下降幅度增加;对保存5年以上的精液需抽样测定功能和品质后再用于生产。To study the effects of cryopreservation( - 196℃)on sperm enzyme's activities in wild yak. The probability of integrated acrosome was detected by giemsa pigmentation. The activity of acrosin was assayed with N - α - benzoyl - DL - arginine-ρ- nitroanilide(BAPNA). The hyaluronidase activity was measured by improved gelatin of substrate membrane, and the acid phosphatase activity was measured by di - sodium phenyl phosphate. The activity of lactate dehydrogenase was assayed by means of enzyme kinetical spectrophotometry. It indicated that the enzyme's activities descended obviously, when the wild yak sperms were cryopreservated. With the cryopreservation time prolonged, the degree of descent ascended.
