肝细胞增殖抑制因子抗血清的制备及其肝内原位表达

摘  要:用90%纯度的肝细胞增殖抑制因子(HPI)制备了效价为1∶32的兔抗鼠HPI抗血清,并检测了HPI在肝内的原位表达。结果表明,正常成年大鼠、兔和豚鼠肝细胞均表达HPI,肝小叶中央带肝细胞表达尤强,非肝细胞无HPI表达。胎鼠和新生鼠肝细胞及非肝细胞无HPI表达。肾内各处结构和细胞也无HPI表达。在肝大部切除后1~2d,肝细胞无HPI表达,4d后呈弱阳性表达并逐渐增强,至10d时达正常成年鼠表达水平。在肝癌发生中,转化肝细胞和肝癌细胞无HPI表达。研究结果提示。he rabbit anti rat HPI (hepatic proliferation inhibitor, HPI) antiserum was prepared with the titer of 1:32 (this 90% purity of HPI was purified in our laboratory) as the first antibody to examine the expression of HPI in livers. The results demonstrated that the HPI expression appeared in the hepatocytes of livers of normal adult rat, rabbit and guinea pig), and that the hepatocytes in centrilobular areas showed much strong HPI expression, whereas there was no HPI expression in the nonparenchymal cells. All the hepatocytes and nonparenchymal cells in fetal and new born rat livers showed no HPI expression, and the different cells in rat kidney also showed no HPI expression. During the first two days after subtotal hepatectomy (SH), the residual hepatocytes displayed no HPI expression, on the 4th day after SH, HPI expression appeared weakly, henceforce, it enhenced gradually; on the 10th day after SH, HPI expression approached the same intensity as normal adult rat livers. In the course of hepatic carcinogenesis, the transformed hepatocytes and hepatoma cell did not express HPI. These results suggested that the HPI may play an important role in the control of the proliferation, differentiation, transformation and carcinogenesis of hepatocytes.
