更年归肾丸对雌性去势大鼠生殖内分泌、免疫功能及自由基代谢的影响

摘　　要：目的：为探讨更年归肾丸临床治疗围绝经期综合征的作用机理，通过药理实验观察该方对雌性去势大鼠生殖内分泌、免疫功能及自由基代谢的影响。方法：将3—4月龄SD雌性大鼠（200g±20g）用手术去除卵巢后分为更年归肾丸中剂量组、高剂量组和妇复春胶囊组，并与去势空白组和假手术组作对照。除去势空白组外，分别灌药20d，然后处死全部大鼠，立即取血测定血清雌二醇（E2）、促卵泡生成激素（FSH）、促黄体生成激素（LH）、白细胞介素2（IL-2）、超氧化物歧化酶（SOD）、谷胱甘肽过氧化物酶（GSH—Px）、丙二醛（MDA）水平等。结果：更年归肾丸能明显提高去势大鼠血清E2、IL-2、SOD水平（P〈0．05）和GSH—Px（P〈0．01）水平，降低MDA含量（P〈0．05）。结论：更年归肾九能在一定程度上提高去势大鼠的E2水平，能明显升高去势大鼠血清IL-2含量，从而增进去势大鼠的细胞免疫功能；能提高血清SOD、GSH—Px水平，降低MDA水平，有利于清除体内自由基，延缓衰老。Objective： To investigate the mechanism of clinical treatment of perimenopausal syndrome of Gengnian Gnishen pill, the effect on reproductive endocrine, immune function and free radical metabolism of female castrated rats were observed by pharmacological experiment. Methods： 3 -4 month -old female SD rats （200 g ±20 g） with ovaries excised were randomized into three groups with treatment of Guishen high dose , middle dose and Fufuchun capsule , which were compared with castrated blank group and pseudo - operated group. All the rats were killed after giving medicine for 20 days except castrated blank group, and Ex, FSH, LH, IL - 2, SOD, GSH - Px and MDA of blood was determined. Result： Gengngian Gnishen pill can obviously increase E2, IL - 2, and SOD （ P 〈 0. 05 ）, GSH - Px level （ P 〈 0. 01 ）, and obviously decrease MDA level （ P 〈 0.05 ） of castrated rats. Conclusion： Gengnian Gnishen pill can obviously increase estrogen level, and obviously increase serum IL - 2 content of castrated rats to improve cell immune function. It can obviously incease serum SOD, GSH - Px level and decrease MDA level , which is beneficial to clear away free radical inside body and retard senility.
