摘  要:【目的】初步探讨白藜芦醇(resveratrol,Res)抗白血病的免疫学机制。【方法】用MTT比色法测定肿瘤细胞生长抑制率,流式细胞术分析其细胞周期。分别采用溶血分光光度法、改良的Mayer法及双抗体夹心ELISA法,检测荷瘤小鼠的免疫功能及细胞因子含量变化。【结果】在一定范围内Res可以时间-剂量依赖的方式抑制白血病L1210细胞增殖、促进其凋亡。Res可促进荷瘤小鼠T细胞增殖、抗体的生成,并且上调IL-2、减少IL-8的分泌水平。【结论】Res可直接抑制白血病L1210细胞增殖,并且通过调节机体免疫应答的水平,协同发挥抗肿瘤作用。[Objective] To discuss preliminarily the immunological mechanism of the anti-leukaemic function of resveratrol (Res) through in vitro or vivo experiments. [Methods] The tumor cell growth repression rate was measuresed by MTT colorimetry, and flow cytometry was used to analyze the cell cycle. The immundogical function and cytokine levels in the serum of the tumor-bearing mice were determined by M T T colorimetry, hemolysis spectrophotography, improved Mayer' s method and ELISA. [Remilts] Res inhibited the of the leukaemia cells growth and enhanced the apoptosis of tumor ceils in time-concentration dependent manners, showing the phenomenon of obvious G0/G1 blocking and apoptotic peak. Res not only improved the function of T, B lymphocytes, but also increased the contents of IL-2 and reduced IL-8. [Conclusion] Res could exert anti-leukaemic efficiency through regulating immune response and secreting cytokines.
