摘  要:目的:建立以RP-HPLC法测定人血浆中齐多夫定浓度的方法。方法:色谱柱为Kromasil C18,流动相为0.5mol·L-1磷酸二氢钾溶液-甲醇(80∶20),流速为1.0mL·min-1,检测波长为267nm,灵敏度为0.002AUFS,柱温为室温。结果:齐多夫定和内标(氟康唑)的保留时间分别为8.1、5.7min;齐多夫定检测浓度在10~2000ng·mL-1范围内与峰面积积分值线性关系良好(r=0.9998),绝对回收率>97%,日内和日间RSD<9.4%;检测限和定量限分别为5、10ng·mL-1。结论:本法适用于齐多夫定血药浓度检测及临床药学研究。OBJECTIVE: To establish a RPHPLC method for the determination of zidovudine concentration in human plasma.METHODS: The determination was performed on a kromasil C18 column, with 0.5mol · L^- 1 potassium dihydrogen phosphate-methanol(80 : 20) as mobile phase,at a flow rate of 1.0mL· min^-1,a detection wavelength of 267nm,a sensitivity of 0. 002AUFS,and under room temperature.RESULTS: The retention time for zidovudine and that for internal standard(fluconazole) was 8.1min and 5.Tmin, respectively. A good linearity was seen with the calibration curves when the detection concentration of zidovudine was within a range of 10-2 000ng · mL^-1 (r = 0.999 8) .The absolute recovery of zidovudine was〉 97%. The intraday and inter-day precision(RSD) was 〈9.4% .The detection limit and quantization limit were 5, 10ng · mL^-1 respectively.CONCLUSIONS: This method can be used for the determination of zidovudine concentration in human plasma and for the study of clinical pharmacy.
