摘 要:以阔瓣含笑、平伐含笑、含笑为材料,对这3个树种的花期进行跟踪观察,并且采用TTC染色法和固体培养基法对它们的新鲜花粉和4℃低温下保存的花粉进行了生活力的测定。结果表明:培养基中蔗糖浓度对花粉萌发具有明显影响,阔瓣含笑花粉的最佳萌发浓度为10%,而平伐含笑和含笑的最佳萌发浓度均为15%;TTC染色法测定的花粉生活力比萌发法高;4℃低温保存条件下,阔瓣含笑花粉的可贮藏时间最短,约40 d后花粉便失去了生活力,而含笑和平伐含笑的花粉贮藏40 d后的萌发率分别为28.77%、32.01%。The pollen viability of fresh pollen and preserved pollen from three species of Michelia were studied with TTC staining method and the solid culture medium method. The results showed that the concentration of sucrose effected on the pollen germinating rate significantly. The germinating rate of fresh pollen of Michelia platypetala reaehed the highest with 10% sucrose, the highest germinating rate of fresh pollen of Michelia cavaleriei and Michelia figo with 15% sucrose. The pollen viability tested by the Trc staining method was higer than that by gemination method. The study of 4 ℃ cold storage showed that the pollen of Michelia platypetala lost viability nearly after 40 days and the pollen of Michelia cavaleriei and Michelia figo retained the viability with 32.01% and 28.77% after 40 days.
