摘  要:目的:探讨氟伐他汀对哮喘气道重建的影响。方法:大鼠气道平滑肌细胞(ASMC)原代培养,MTT法观察细胞增殖状况,流式细胞仪检测细胞周期变化;构建大鼠慢性哮喘模型,随机分为4组,即:空白对照组(A组)、哮喘组(B组)、氟伐他汀组(C组)、地塞米松组(D组)。测量气道壁厚度和气道内外径比值,以及免疫组化法半定量法检测PCNA、Ⅲ型胶原表达含量。结果:氟伐他汀对于由组胺、血清刺激增生的ASMC有明显的抑制作用,细胞分裂多数停滞于G1期。大鼠哮喘模型中,C组的气道壁厚度、PCNA、Ⅲ型胶原表达明显低于B组,差别均具有统计学意义(P<0.05),C组气道壁厚度、PCNA、Ⅲ型胶原表达高于D组,差异均有统计学意义(P<0.05)。结论:氟伐他汀对哮喘气道重建具有抑制作用。Objective:To investigate the effects of fluvastatin(Flu), a hydroxymethylglutaryl - CoA(HMG - CoA) reduease inhibitor, on airway remodeling in asthma. Methods, Airway smooth muscle cells(ASMC) in rats were cultured in vitro. The proliferation of ASMC was detected with MTT and the cell cycle was detected by flow cytometer. An asthmatic model of rat was established. 32 SD rats were divided randomly into four groups with eight rats in each group, i. e. normal group(A), model group(B), Flu treated group(C) and dexamethasone treated group(D). The changes of collagen type Ⅲ and proliferating cell nuclear antigen(PCNA) content in the airway wall were detected with immunohistochemical method, the thickness of smooth muscle and the ratio of inner diameter to outer one were measured before and after the treatment. Results:The stimulative effects of histamine and serum on the proliferation of ASMC were prevented by Flu. Flu blocked significantly the cell cycle progression in G1 phase The thickness of smooth muscle and collagen type Ⅲ and PCNA in the airway wall in group C were significantly lower than those in group B(P〈0. 05) and significantly higher than those in group D(P〈0. 05), Conclusion:Flu could inhibit the airway remodeling in asthma.
