摘  要:目的探讨戊四氮(PTZ)反复点燃对发育鼠海马神经元发生的影响。方法10日龄Wistar大鼠48只,随机分为PTZ组和对照组,每组各24只。5-溴,2-脱氧尿嘧啶核苷(BrdU)标记新生细胞。二组分别于模型制备完成后24 h、3 d及1个月时各处死8只大鼠;观察其惊厥行为学改变,海马区细胞形态、细胞计数,免疫组织化学法检测海马各区核因子-κB(NF-κB)表达及BrdU阳性细胞数,Timm染色法观察苔藓纤维发芽。结果1.模型制备完成后24 h,PTZ组海马CA1、CA3、DG区NF-κB表达均显著高于对照组(t=9.081,12.044,11.002 Pa<0.01);2.模型制备完成后3 d,PTZ组齿状回(DG)颗粒细胞数及BrdU阳性细胞数明显高于对照组(Pa<0.05),CA1、CA3及门区无明显神经元变性坏死现象;3.模型制备完成后1个月,PTZ组有苔藓纤维发芽,对照组无明显苔藓纤维发芽。结论PTZ反复点燃可诱发发育脑海马神经元的发生。Objective To investigate the influence of pentetrazole (PTZ) repeated kindling on hippocampal neurogenesis in developing rats. Methods Ten - day - old Wistar rats ( n = 48 ) were prepared for epilepsy model. They were divided into PTZ group and control group at random, each group had 24 rats. The brornedeoxy uridimel BrdU was utilized to label newborn ceils. The rats were killed in 24 hours, 3 days and 1 month respectively after the model completed. The behavioral changes, the morphology and neurons counts in hippocampus were observed, the expression of nuclear factor - κB (NF - κB ), BrdU - immunoreactive cells in hippocampu was detected with immunohistochemistry, and the mossy fiber sprouting were observed. Results 1. Twenty - four hours after the model completed in PTZ group, the NF - κB expressions in hippocampu was significantly higher than that in control group( t = 9. 081,12. 044,11. 002 Pa 〈 0.01 ). 2. Three days after the model completed, in PTZ group, the dentate gyrus ceils count and BrdU -immunoreactive ceils count were significantly higher than those in control group( Pa 〈 0.05 ), while the CA3, CA1 and hilar ceils count had no significant difference compared with that in control group. 3. One month after the model completed, mossy fiber sprouting could be observed in PTZ group, while in control group, there was no significant mossy tiber sprouting. Conclusion PTZ repeated kindling can induce hippocampal neurogenesis in developing rats.
