摘  要:目的:探讨包含有重复序列TTAGGG的寡聚脱氧核糖核苷酸(ODNA151)对不稳定型心绞痛(UAP)患者外周血辅助性T(Th)细胞功能平衡的影响。方法:分离16例UAP患者、10例稳定型心绞痛(SAP)患者和12例胸痛综合征(CPS)患者外周血单个核细胞,培养后应用ELISA法检测Th细胞功能。对UAP患者,在培养体系中加入不同浓度的ODNA151或对照的ODN1612,采用ELISA法测定上清中Th1型细胞因子IFN-γ和Th2型细胞因子IL-4的浓度,评价其对Th亚群功能的影响。结果:与SAP和CPS组比较,UAP组患者Th1细胞功能明显增强。ODNA151抑制UAP患者淋巴细胞培养上清的IFN-γ浓度,并升高IL-4的浓度,降低IFN-γ与IL-4的比值;而对照的ODN1612对细胞因子水平无影响。结论:UAP患者Th1细胞功能明显增强,ODNA151对UAP患者淋巴细胞的Th1/Th2平衡有调节作用,既抑制Th1分化又促进Th2分化。Objective:To explore the effect of oligodeoxynucleotides (TTAGGG) on peripheral T helper (Th) lymphocytes in patients with unstable angina (UA). Method: The Th cell-functions were detected in 16 patients with UA, 10 patients with stable angina (SA) and 12 patients with chest pain syndrome (CPS) by ELISA. Peripheral blood monocytes from UA patients were cultured with phytohemagglutinin (PHA) and different concentrations of (1, 5, 10 μmol/L)ODNA151 or ODN1612 (10 μmol/L) for 72 hours, then IFN-γ and IL-4 concentrations in supernatant were detected by ELISA. Result: The Thl cell-functions were found to be significantly higher in patients with UA than those with SA and CPS. ODNA151 but not ODN1612 significantly inhibited IFN-γ and increased IL-4 (P〈0.0 1), which accordingly down-regulated ratios of IFN-γ/IL-4. Conclusion:ODNA151 regulated differentiation of Th lymphocytes in patients with UA and skewing Th1/Th2 balance toward Th2 type cytokine production.
