摘 要:目的制备、鉴定基于肿瘤抗原survivin的“模拟病毒”瘤苗。方法在特定的条件下将阳离子肽和DNA制备成模拟病毒颗粒样的结构,并以扫描电镜、DNA阻滞试验、DNaseⅠ保护试验和免疫印迹(Western blotting)对该疫苗进行制备鉴定。结果在NaCl浓度为87.5 mmol/L,电荷比为2的制备条件下,该疫苗能形成颗粒,并可有效介导瘤苗所携带基因的表达。结论该研究成功制备了基于肿瘤抗原survivin的“模拟病毒”瘤苗,为新型的肿瘤疫苗的设计奠定了基础。Objective To prepare and identify the mimovirus tumor vaccine based on survivin. Methods Under specific condition, peptide and DNA were condensed into like-virus particle, and the preparing parameters were optimized by assays of DNA gel retardation, DNase I protection and identified by Western blotting. Antigen presentation was also studied by RMA-S/Kd cells. Results The vaccine was condensed into homogeneous particles at charge ratio of 2 in 87.5 mmol/L NaCl concentration, which could effectively mediate expression of the genes carried by vaccine particles. Conclusion The mimovirus tumor vaccine based on survivin was prepared successfully, providing a good foundation for the design of new tumor vaccine.
