摘 要:用RAPD技术对家蚕和柞蚕进行基因组DNA分析,研究和比较家蚕和柞蚕之间的遗传多态性,确定家蚕和柞蚕的遗传差异。该实验中共使用了6种随机引物:5'-GACCGCTTGT-3’,5'-CAGGCCCTTC-3,5'-TGACGGCGGT-3’,5'-CAGCACTGAC-3’,5'-CAGCACTGAC-3',5'-CAACA3C,CAGA-3',扩增出60条清晰的条带;单个随机引物的扩增条带数在8—11条;所扩增的基因组DNA条带的分子量在0.3~3.3kb。计算结果得出家蚕与柞蚕的遗传距离较大,平均距离约为0.639。实验还发现,在等量组织、同条件下抽提DNA时,从丝腺中抽提出的DNA量最大,从中肠和脂肪中抽提出的DNA量相对较少。Random Amplified Polymorphic DNA (RAPD) was used to analyze the genetic diversity between Antheraea pemyi and Bombyx mori, The result showed that six arbitrary primers which were used in this experiment could be amplified clearly. And a total of about sixty patterns were obtained. Each primer gave eight to ten hands.The genetic distance between Bombyx mori and Antheraea pemyi was 0.639. The result also showed that on the same conditions and with the same content of tissue,the content of DNA in silk gland was the most. The content of DNA in midgut and fat body was less than silk gland.
