摘 要:应用单细胞凝胶电泳技术(SCGE),对冷冻解冻后猪精子DNA的损伤情况进行研究。结果表明,冷冻解冻会对猪精子DNA造成损伤,其彗星率与鲜精相比差异显著(P〈0.05);各组冻精之间,甘油添加量为2%~3%(v/v)时,冷冻对猪精子DNA造成的损伤最低,彗星率均低于其余各组(P〈0.05)。Boar sperm cryopreservation is an important method for germplasm conservation. However, the method may cause the change of DNA structure and component. The change of DNA structure may damage the boar sperm quality and result in defects in its descendant. Therefore,how to keep the sperm intact and how to avoid the damage caused by cryopreservation become an important issue. Thus, the effect of cryopreservation on the DNA damage of boar sperm was studied and the method of single cell gel electrophoresis(SCGE) was a simple and valid method to examine the damage of the sperms. And the degree of the DNA damage was estimated with the comet tail pattern formed by the DNA nuclei movement in the electricity field. SCGE was used to study the DNA damage of cryopreservation boar sperm. The result indicated that cryopreservation can cause boar sperm DNA damage and compared with the fresh sperm it had remarkable difference( P 〈0.05), the frozening caused lowest DNA damage than others when the glycerol with the concentration of 2% to 3% was suppled in experiment group( P 〈0.05).
