清肝益气降压方对自发性高血压大鼠肾内小动脉血管紧张素Ⅱ1型受体蛋白表达的影响

摘　　要：目的:观察清肝益气降压方对自发性高血压大鼠(SHR)肾内小动脉血管紧张素Ⅱ1型受体蛋白表达的影响,探讨本方的降压机制。方法:30只雄性SHR大鼠随机分为模型对照组、阳性对照组、清肝益气组、清肝泻火组和疏肝抑火组,连续给药6周,以Wistar大鼠作空白对照。测定各组大鼠血压、血管紧张素Ⅱ水平,用原位杂交技术检测血管紧张素Ⅱ1型受体(AT1R)蛋白的表达。结果:清肝益气降压方有确切的降压作用,清肝益气组与模型对照组相比血压明显下降(P<0.05);能明显降低AngⅡ水平(P<0.05),能抑制SHR大鼠肾内小动脉AT1R蛋白的表达(P<0.05)。结论:清肝益气降压方具有降压作用,提示通过抑制AT1R蛋白的表达,降低AngⅡ水平,可能是该方的降压机制。Objective：To investigate the effect of antihypertenive formula of clearing liver fire and supplementing qi on the expression of AT1R protein of the arteriolar in kidney of spontaneously hypertensive rat（SHR）, and to explore the antihypertensive mechanisms. Methods：Thirty male spontaneously hypertensive rats were randomly divided into five groups（ model group, positive control group, clearing liver fire and supplementing qi group, clearing liver fire group, soothing the liver and restraining liver fire group）, and were given the drugs for six weeks, and addtional six Wistar rats were regarded as normol group. Blood pressure and Ang Ⅱ in plasma were detected, and the effect on the expression of AT1R protein of the arteriolar in kidney were detected by in situ hybridization. Results： The formula showed antihypertensive effect, with marked blood pressure drop compared with the model （ P 〈 O. 05） ; and restrained the Ang Ⅱ level and the expression of AT1R protein of the arteriolar in kidney（ P 〈 O. 05）. Conclusions：The formula of clearing liver fire and supplementing qi has good antihypertensive effect. The antihypertensive mechanism might be that the formula can restrain the expresson of AT1R protein of the arteriolar in kidney and reduce the level of Ang Ⅱ in plasma.
