摘  要:目的:观察复方接骨中药含药血清培养诱导的骨髓间充质干细胞(MSC)与去抗原牛松质骨(BCB)复合体在大鼠体内的成骨作用。方法:取3月龄的雄性SD大鼠32只随机分为实验组与对照组,实验组将含药血清培养诱导的骨髓MSC-BCB复合体种植于大鼠皮下,对照组将单纯BCB直接种植于大鼠皮下。实验组与对照组分别于术后14和28 d各取出8个植入物。取每个植入物的一半于4%多聚甲醛溶液固定,石蜡包埋后切片,行HE染色,光镜下观察。另一半定量后匀浆,麦胚凝集沉淀法测定骨源性碱性磷酸酶(BALP)活性。结果:实验组植入物术后14 d光镜下见大量成骨细胞附于BCB骨小梁表面,术后28 d大量软骨腺泡形成。对照组成骨细胞数量较少,骨小梁间隙仅见纤维组织,未见新骨发生。同期实验组BALP活性显著高于对照组(P<0.05)。结论:复方接骨中药含药血清体外诱导的骨髓MSC-BCB复合体具有体内成骨作用。Objective :To observe the effects of mesenehymal stem cell(MSC) and antigen-extracted bovine cancellous bone ( BCB ) complex cultured and induced by serum containing Traditional Chinese Compound Bone Healing Recipe (TCCBHR) on osteogenesis in rats. Methods:32 three-month-old male SD rats were divided into the control and experimental groups. In the experimental group,the MSC-BCB complex was implanted subeutaneouly. In the control group,only. BCB was implanted subeutaneouly. Then 8 planted things were taken from the control and experimental groups respectively 14 days and 28 days after operations. The half of each implant was fixed by 4% paraformaldehyde (PFA) solution,sliced after being embedded by paraffine, then stained by HE and observed under light microscope.The other haft of each implant was weighted and grinded,and the activity of BALP was determined by wheat-embryo curdling and deposition method. Results: In the experimental group,under light microscope, there were a great number of osteoblasts and the cells adhered to the surface of bone's girder at the end of the 2nd week. At the end of the 4th week ,a great number of cartilage gland alveolus were generated, but in the control group, the osteoblasts number was smaller than that of the experimental group and the surface of trabeeular bone had only fibrous tissue appearance at the bone girder clearance,and no formation of new bone were found. The Bone alkaline phosphatase (BALP) activity of the experimental group was obviously greater than that of the control group at the end of the 2nd week. Conclusion:The MSC-BCB complex euhured and induced by serum containing TCCBHR possesses osteogenesis in rats.
