杏丁注射液对糖尿病大鼠肾脏的保护作用

摘　　要：目的:探讨杏丁注射液对糖尿病(DM)大鼠肾脏病变的保护作用和机制。方法:腹腔注射链脲菌素60mg/kg制成2型糖尿病大鼠模型,以72小时后空腹血糖>16.7mmol/L为糖尿病模型标志。动物随机分为正常对照组、DM模型组、杏丁组。第12周末分别测定尿白蛋白、血糖、甘油三酯、胆固醇、尿素氮和肌酐,测定肾组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性、一氧化氮(NO)、一氧化氮合成酶(NOS)及肾脏肥大指数。结果:(1)DM模型组与正常对照组相比,尿白蛋白、BUN、Scr明显升高,肾脏肥大指数增加;肾组织GSH-Px、SOD和NO、NOS活性降低(P<0.01);光镜和电镜下肾组织可见相应的病理改变。(2)杏丁组与DM模型组相比,大鼠24h尿白蛋白量明显下降(P<0.01),肾组织GSH-Px、SOD、NO、NOS活性增加(P<0.05),肾脏肥大指数降低(P<0.01),同时肾脏病理改变亦减轻。结论:杏丁注射液能改善和调节DM大鼠肾组织过氧化反应指标,对DM肾脏损害有保护作用。Objective： To investigate the protective effect of Xinding Injection on kidney in diabetic rats. Methods： Diabetic rats were induced by intraperitoneal Streptozotocin injection（60mg/kg） at one dose and measured the blood fasting sugar after 72 hours .Consistency of blood fasting sugar in the diabetic model rats was onto 16.7mmol/L. The rats were randomly divided into 3 groups： the normal control group （NC）, the diabetic model group （DM） and the Xinding -treated diabetic group （XD）. At the end of 12weeks, glucose, triglyceride （TG）, total cholesterol （TC）, urea nitrogen, creatinine level of plasma and 24 urinary albumin excretion were detected. The relevant indexes in tissue fluid of kidney and the kidney hypertrophy index were measured. Results： 1. The 24-hour urinary protein, BUN, Scr and the kidney hypertrophy index were higher in Group DM than that in Group NC. The activity of GSH-Px, SOD, NO and NOS in tissue fluid of kidney were lower in Group DM than that in Group NC （P〈0.01）.The corresponding pathological changes of renal tissue could be obviously observed under light microscopy and electron microscopy in Group DM. 2. Xinding Injection could reduce 24 hours urine protein, kidney hypertrophy index and significantly increase the activity of of GSH -Px, SOD, NO and NOS in tissue fluid of kidney, compared with the DM group （P〈0.05）. The pathological changes in Group XD were less obvious than those in Group DM. Conclusion： Xinding injection can perfect the relevant indexes in tissue fluid of diabetic rats kidney. Xinding injection has some protective effect on kidney of diabetic rat.
