高效液相色谱法测定地红霉素肠溶片中地红霉素的含量

摘　　要：目的建立HPLC法测定地红霉素在其肠溶片中的含量。方法采用Hypersil BDS C18柱（200mm×4．6mm，5μm）；流动相为乙腈-磷酸盐缓冲液（取1．41g磷酸二氢钾与6．91g磷酸氢二钾，加水1000mL溶解后，过滤，即得）-甲醇（44：37：19）；检测波长为205nm；柱温40℃。结果地红霉素在0．8～4．7mg·mL^-1浓度与峰面积呈良好的线性关系（r=0．9999），平均回收率为99．5％，RSD为0．31％（n=9）。结论方法简便、快速、灵敏、准确。Objective To establish an HPLC method for determining dirithromycin in its enteric-coated tablets. Methods The Hypersil BDS C18 Column （200 min×4. 6 mm, 5 μm） was used with the mobile phase of acetonitrile phosphate buffer （ 1.41 g potassium dihydrogenphosphate and 6.91 g potassium phosphate dibasic dissolve and filtrate in 1 000 mL water ） -methanol （44 ： 37 ：19） at the detection wavelength of 205 nm. The column temperature was 40 ℃. Results Good linearity was found within 0. 8-4.7 mg · mL^- 1 （r=0. 999 9）, and the average recovery was 99.5% with RSD of 0.31% （n=9）. Conclusion The method is simple, rapid, sensitive and accurate.
