摘 要:目的建立测定当归苦参丸中苦参碱和氧化苦参碱含量的高效液相色谱法。方法采用Phenemonex C18(250mm×4.6mm,5μm)色谱柱,流动相:乙腈-0.1%三氟醋酸溶液(5:95),流速:1.0mL·min^-1,检测波长:208nm,柱温:35℃。结果苦参碱和氧化苦参碱线性范围分别为0.0512~0.3584μg(r=0.9990)和0.1924~1.3468μg(r=0.9999);平均回收率分别为98.73%,100.14%(n=6)。结论该方法操作简单,重现性好,检测灵敏度、准确度高,且样品分离效果好。适用于当归苦参丸中苦参碱和氧化苦参碱的含量测定。Objective To establish an HPLC method for the determination of matrine and oxymatrine in Dangguikushen pills. Methods The column was Phenomenex Gimini C18 (250 mm×4.6 mm, 5μm). The mobile phase was acetonitrile -0.1% trifluoroacetic acid (5 : 95) with a flow rate of 1.0 mL · min^-1. The detection wavelength was 208 nm. The column temperature was 35℃. Results The linear ranges of matrine and oxymatrine were 0. 051 2-0. 358 4μg and 0. 192 4-1. 346 8μg. The average recoveries were 98.73% and 100. 14%. Conclusion The method is simple and accurate, and can be used for the determination of matrine and oxymatrine in Dangguikushen pills.
