摘  要:目的研究大鼠骨髓来源树突状细胞(DC)的培养扩增方法及用肿瘤细胞裂解物致敏的树突状细胞(TAA/DC)的免疫学特性。方法用含重组大鼠粒细胞-巨噬细胞集落刺激因子(rrGM-CSF)、重组大鼠白细胞介素-4(rrIL-4)和重组大鼠肿瘤坏死因子-α(rrTNF-α)的培养体系将近交系大鼠骨髓细胞诱导扩增为DC;用大鼠卵巢癌细胞的肿瘤细胞裂解物(TAA)致敏未成熟DC,获得TAA/DC;用流式细胞术分析DC、TAA/DC细胞的免疫表型。结果含rrGM-CSFr、rIL-4和rrTNF-α的培养体系可在体外诱导大鼠骨髓单个核细胞分化为成熟的DC;TAA可促进DC的分化成熟,TAA/DC的细胞免疫表型显著增强。结论大鼠骨髓细胞可在体外诱导为成熟的DC;负载肿瘤全抗原的DC表达与抗原提呈相关的分子增高,提示其抗原提呈能力增强。Objective To study the culture method of dendritic cells (DCs) derived from rat bone marrow and to analyze the immune characteristics of DCs sensitized with tumor associated antigen (TAA/DC). Methods The culture system including granulocyte-macrophage colony-stimulating factor (GM-CSF), interleukin-4 (rrIL-4) and tumor necrosis factor-α (TNF-α) was used to induce and expand DCs;Immature DCs were pulsed with tumor associated antigen (TAA) to obtain TAA/DCs. The immunophenotypes of DCs and TAA/DCs were analyzed by using flow cytometry. Results The culture system containing GMCSF, rrIL-4 and TNF-α could induce the differentiation of monocytes derived from rat bone marrow into mature DCs in vitro. TAA could promote the differentiation and mature of DCs. The immunophenotypes of TAA/DCs were raised remarkably. Conclusion The rat bone marrow cells can be induced in vitro to differentiate into mature DCs;TAA/DCs can load the total antigen of tumor cell, and the capability of antigen-presenting is improved.
