摘 要:目的探讨高浓度氧暴露对早产大鼠肺组织中还原型烟酰胺腺嘌呤二核苷酸脱氢酶亚单位1(ND1)表达的影响。方法早产SD新生大鼠生后1 d随机分为高氧组和空气组,高氧组持续暴露于≥85%氧气中,空气组置于同一室内常压空气中。分别取两组大鼠高氧或空气暴露后1、4、7、10和14 d肺组织标本,采用免疫组织化学和Western blot方法检测ND1蛋白的表达,半定量逆转录聚合酶链反应(RT-PCR)检测ND1 mRNA的表达。结果①高氧组和空气组各时间点均有ND1蛋白表达;与空气组相比,高氧组在1、4和7 d ND1蛋白表达增高,随后其表达下降。②与同时间点空气组相比,高氧暴露后1、4和7 d ND1 mRNA表达显著增高(P<0.05),其后ND1 mRNA表达逐渐下降,10 d和14d时低于空气组,但两者相比差异无显著性意义(P>0.05)。③ND1蛋白的表达变化趋势与ND1 mRNA的表达变化趋势一致。结论高氧导致早产大鼠肺组织ND1表达改变,提示ND1可能参与了高氧肺损伤的病理生理过程。Objective To investigate the effects of hyperoxia on the expression of NADH dehydrogenase subunit 1 (ND1) in premature rat lung. Methods Premature Sprague-Drawley rats were randomly assigned to hyperoxia group and air group at first day after birth. The rats in hyperoxia group were persistently exposed to ≥85% oxygen and those in air group to room air. The lung tissues of both groups were collected at 1st, 4th, 7th, 10th and 14th day after being exposed to hyperoxia or air. The expression of ND1 protein was detected by immunohistochemistry and Western-blot. The expression of ND1 mRNA was detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). Results ND1 protein was expressed both in hyperoxia group and air group, As compared with air group, the expression of ND1 protein was increased at 1st, 4th and 7th day and decreased at 10th and 14th day. As compared with air group, the ND1 mRNA level in hyperoxia group was significantly increased (P〈0.05) at 1st, 4th and 7th day, then gradually decreased, lower than air group at 10th and 14th day, but there was no significant difference (P〉0.05). The expression tendency of ND1 protein was consistent with ND1 mRNA. Conclusion Hyperoxia induced the changed expression of ND1 in premature rat lung, indicating that ND1 may participate in the physiological and pathological processes of hyperoxia-induced lung injury.
