滋阴益气活血解毒组方对MKR转基因2型糖尿病小鼠糖代谢的影响

摘　　要：目的观察滋阴益气活血解毒组方-左归复方干预治疗对骨骼肌胰岛素样生长因子-1及胰岛素双受体功能缺失(MKR)转基因2型糖尿病小鼠的糖代谢的影响。方法由美国国立卫生研究院(NIH)授权使用引进的MKR鼠,经自然交配后繁殖的子代作为研究对象。MKR小鼠经繁殖鉴定后,以左归复方给药30d进行干预治疗。结果MKR鼠所有子代鼠均会出现长度为330bp的DNA片断,表明该MKR鼠后代能稳定遗传。新生MKR鼠自出生3周开始,即出现显著的血糖增高,5周以后血糖则较稳定地维持在高水平。与对照组小鼠同期的血糖比较,差异具有非常显著性意义(P<0.01)。而左归复方给药30d进行干预治疗后,其高剂量具有显著的改善MKR鼠糖耐量异常和降低空腹血糖的作用(P<0.05,P<0.01)。结论我们成功地引进了MKR模型,MKR鼠是目前一种非常良好的2型糖尿病的动物模型。以滋阴益气活血解毒立法化裁组方的左归复方具有显著的改善MKR鼠糖耐量异常和降低空腹血糖的作用。Objective To establish the MKR mouse, a model in which expression of a dominant-negative IGF-I receptor in the skeletal muscle leads to systemic insulin resistance and diabetes, and examine changes in the circulating glucose level treated by invigorating Yin and Qi and detoxicating and promoting blood flow, also namely Zuogui recipe, in the MKR mice. Methods MKR mice were genotyped by a specific PCR reaction using tail DNA. These mice were followed on blood glucose level and body weight once a week from 1 to 16 weeks of age. And the MKR mice were treated by ZuoGui recipes for 30days. Results The offspring of MKR mice exhibited stable transgeni（; hereditary. These mice were hyperglycaemic from 3 weeks of age onward; the body weight increase slowed down after 4 month. After treating of ZuoGui recipe, the MKR mice were significantly improved glucose intolerance and decreased hyperglycemia. Conclusion We have estabolished the MKR type 2 diabetic model. And ZuoGui recipe could improve glucose intolerance and decrease hyperglycemia in the MKR mice.
