摘  要:目的研究补阳还五汤及其有效部位生物碱、苷对大鼠脑缺血再灌注后白细胞介素1β(IL-1)β及其相关因子mRNA表达的影响。方法采用颈内动脉线栓法建立大鼠MACO再灌注模型,以逆转录-聚合酶链式反应(RT-PCR)测定缺血脑组织IL-1β、IL-1R1、IL-1ra、ICAM-1 mRNA的表达。结果脑缺血2h再灌注22h后,模型组IL-1β、IL-1R1、IL-1ra和ICAM-1 mRNA表达较假手术组均显著增强(P<0.05和P<0.01)。补阳还五汤组IL-1β表达显著高于假手术组(P<0.05),而生物碱组和苷组较模型组有所降低;补阳还五汤组IL-1R1表达较模型组显著降低(P<0.05),而生物碱组和苷组IL-1R1表达均显著高于假手术组(P<0.05),与模型组接近。补阳还五汤组、生物碱组和苷组IL-1ra表达均显著高于假手术组(P<0.05和P<0.01)。补阳还五汤组、生物碱组、苷组ICAM-1表达低于模型组(P<0.05)。结论补阳还五汤及其有效部位生物碱、苷通过抑制IL-1β、IL-1R1I、CAM-1 mRNA的表达,从而减少炎症因子及炎症介质的表达,对缺血脑组织起保护作用。各药对脑缺血后IL-1ra表达无影响。Objective To investigate the effects of Buyang Huanwu Decoction (BHD) and two kinds of fractions extracted from BHD on the expression of IL-1β and associate factor mRNA after cerebral ischemia reperfusion in rats. Methods The model of cerebral ischemia reperfusion was induced by occlusion of the middle cerebral artery in rats, via surgical insertion (and subsequent removal) of a thread into the internal carotid artery, and the expression of IL-1β, IL-1R1, IL-1ra and ICAM-1 mRNA in cerebral ischemic tissues was assessed by reverse transcription-polymerase chain reaction(RT-PCR). Results The expression of IL-1β,IL- 1R1,IL-1ra and ICAM-1 mRNA in cerebral tissues was significantly increased after 22 hours of reperfusion followed 2 hours of cerebral ischemia in model group (P〈0.05 or P〈0.01), and the expression of IL-1β in BHD group was remarkably increased compared with the sham-operated group (P〈0.05). The expression was reduced in alkaloid group and glycoside group.Compared with model group, BHD decreased the expression of IL-1R1 remarkably in brain tissue (P〈0.05). Compared with the sham-operated group, the expression of IL-1R1 in alkaloid group and glycoside group was enhanced approached to the model group. The expression of IL-1ra was enhanced in BHD group, alkaloid group and glycoside group (P〈0.05 or P〈 0.01). The expression of ICAM-1 in BHD, alkaloid and glycoside groups were decreased(P〈 0.05). Conclusion BHD, alkaloid and glycoside can inhibit the expression of IL-1β, IL- 1R1, IL-1ra and ICAM-1 mRNA after cerebral ischemia reperfusion injury, reduce the expression of inflammation factor and medium, and protect the cerebral ischemic tissues. BHD, alkaloid and glycoside are not effective for expression of IL-lra after cerebral ischemia.
