摘  要:目的动态观察急性肺血栓栓塞症(PTE)中环氧合酶-2(COX-2)在肺内的表达及血浆中血栓烷素B2(TXB2)和6-酮-前列腺素F1α(6-K-PGF1α)的变化,并探讨当归和川芎嗪对它们的影响。方法将大鼠分为对照组、PTE模型组、当归治疗组及川芎嗪治疗组,采用自体血栓回输法建立PTE模型。免疫组化法检测大鼠肺内COX-2蛋白的表达水平,放射免疫法测定血浆中TXB2和6-K-PGF1α的浓度变化。结果PTE模型组肺组织炎症损伤明显,当归及川芎嗪治疗组损伤减轻。免疫组化检测显示7d后模型组COX-2蛋白表达明显高于对照组(P<0.05)。放免法检测显示TXB2与6-K-PGF1α血浆浓度之比(T/P)模型组明显高于明显高于对照组(P<0.05)。结论当归及川芎嗪治疗可明显减少PTE大鼠COX-2的表达,减轻炎症反应。Objective To study the expression levels of cyclooxygenase-2(COX-2) in the lung tissue, the concentrations of thrornboxane B2(TXB2) and 6-keto- prostag1αndin F1α(6-K-PGF1α) in the p1αsma in acute PTE, and to explore the effects of angelica and ligustrazine on their expressions. Methods Rats were dividered into b1αnk group, PTE model group, angelica and Ligustrazine therapy group. The PTE model was established by intravenous injection of autologous blood clots. The pathological changes of the lungs were examined with light microscope. By using immunohistochemistry, the expression levels of COX-2 in lung tissue were examined. The p1αsma's concentrations of TXB2 and 6-K-PGF1α were examined by radioimmunity assay. Results Severe lung injury in the PTE group, but mild in the angelica and ligustrazine group. After seven days, the differences of COX-2 in the lung tissue and the rate of TXB2 and 6-K-PGF1α's concentrations (T/P) in the p1αsma between the b1αnk group and the PTE group was significant (P〈0. 05). Conclusion Angelica and ligustrazine can reduce the expression levels of COX-2 in acute PTE. Traditional Chinese medicine should be considered in acute PTE therapy.
