摘 要:目的研究机采血小板常温保存过程中代谢及体外功能的变化规律。方法通过对15U保存0~9d机采血小板的血气指标、葡萄糖、乳酸、乳酸脱氢酶、血小板膜表面CD62p表达率、凝血酶激活后CD62p再表达水平、血小板低渗休克反应进行系统监测,分析机采血小板体外保存过程中代谢指标与体外功能的关系。结果血小板在体外保存9d的过程中,乳酸与H+逐渐堆积,pH值随之下降;血小板低渗休克反应在7d时开始明显下降;CD62p表达率随保存天数增加而逐渐增高,凝血酶激活后CD62p再表达率逐步下降;血气、葡萄糖、乳酸等代谢指标与血小板低渗休克反应、CD62p的表达率及再表达率具有显著相关性。结论对代谢指标的监测可以间接反映血小板体外功能的变化,从而对血小板保存质量作出系统评价。Objective To investigate the orderliness of metabolism and function in vitro during the storage of plateletapheresis at 22℃ and analyze the correlation between them. Methods We studied blood gases,glucose concentration,lactate concentration and LDH concentration,CD62p expression rate,CD62p anew expression rate activated by human thrombin,and hypotonic shock response (HSR) of 15 units of plateletapheresis during a 9-day storage,and analyzed the correlation between the metabolism and the function in vitro of plateletapheresis. Results Lactate and H^+ were accumulated in plasm,and pH value gradually decreased with the consumption of oxygen ;HSR remarkably decreased from day 7 ;CD62p expression incresed,and CD62p anew expression decreased with the prolonging of storage time. There were significant correlations between the metabolic parameters (blood gases, glucose concentration, lactate concentration) and the functional characteristics (HSR,CD62p expression,CD62p anew expression) in vitro during the storage of plateletapheresis. Conclusion We can know the change of plateletapheresis function in vitro by detecting the metabolic parameters of plateletapheresis during storage,accordingly evaluate platelet quality roundly.
