摘  要:目的探讨MDV(Marek's Disease Virus马立克氏病毒)抗独特型抗体(Ab2)模拟抗原诱导机体产生抗MDV感染的能力。方法MDV抗独特型抗体(7A12、6E5)与转移因子制备成抗体苗,对17日龄胚和1日龄雏鸡分别接种,设火鸡疱疹病毒疫苗(HVT)和非免疫空白对照。分别于30 d;60 d;120 d采血。30日龄后用马立克氏病毒(MDV)标准Ⅰ型强毒-京-1毒行腹腔攻击,监测被免雏鸡免疫系统-淋巴器官重量、循环血淋巴细胞数量、病理组织学变化、孵化率以及免疫抑制性反应。结果免疫攻毒120 d后,MDV抗独特型抗体苗组脾脏增重,肝脏未见淋巴细胞浸润,抗MDV感染保护率为88%,未见出现大肠杆菌的继发感染。HVT苗组脾脏减轻,循环的血淋巴细胞数量下降、出现病理损伤,抗感染保护率为74%。MDV感染组脾脏的重量及循环血中淋巴细胞的数量减少,大肠杆菌感染的敏感性增加。结论MDV抗独特型抗体免疫能诱导机体产生抗MDV感染保护。Objective To study the protective efficacy of anti -idiotypic antibody (Ab2) that simulates the antigen to induce the immunity against the infection of Marek's disease virus (MDV) in chicken. Methods The specific -pathogen free (SPF) chickens were vaccinated with Ab2, which was mixed with transferring factors by intraembryonical - subcutaneous way and simple subcutaneous way at the embryo day ( ED ) 17 and day 1 post hatch, respectively. Then the blood was sampled at day 30, 60 and 120, respectively. The vaccine against herpesvirus of turkey (HVT) was set as the positive control and non - vaccinated as the negative one. Thirty- day old chickens vaccinated with Ab2 were challenged with virulent MDV type I at day 30. The weight of lymph organs, the number of lymphocyte in the circulation, histopathological changes, hatch rate and anti - immunosuppressive reaction with infection of E. coli at the day 30, 60 and 120 were measured. Resuits Neither infiltrates of lymphocytes nor histopathological changes in lymphoid tissues were observed after Ab2 vaccination. Protection rate against MDV in the chickens at day 120 was 88% without any occurrence of succeeding infection of E. coli compared with the negative and positive control( P 〈 0.05 ). However, after the vaccination with the vaccine against HVT both the weight of lymph organs and the number of lymphocytes were decreased significantly in comparison with the control. Additionally the histopathological changed was also observed in the spleen and liver. The protection rate against MDV was 74% at day 120. In the MDV infected chickens the weight of spleen and the number of lymphocytes was reduced with the augmented sensitivity to infect E. coli. Conclusions Immunization of Ab2 can produce the protective immunity against MDV infection in chickens.
