金黄色葡萄球菌L型对肿瘤组织和培养细胞影响的研究

摘　　要：目的探讨金黄色葡萄球菌(金葡菌)L型感染对C57小鼠肿瘤组织和体外培养猴肾细胞的影响。方法用金葡菌L型感染C57小鼠及猴肾细胞,再用革兰氏染色、免疫组化染色和聚合酶链反应(PCR)检测C57小鼠肝组织及诱发的小鼠肿瘤组织中金葡菌L型阳性率,并观察感染后猴肾细胞的病理形态学变化。结果C57小鼠肝组织及肿瘤组织中金葡菌L型感染率分别为0%(0/10)和70%(7/10),L型抗原表达率分别为0%(0/10)和70%(7/10);金葡菌L型DNA阳性表达率分别为0%(0/10例)和70%(7/10例)。猴肾细胞感染L型原生小体后体外培养48h部分出现空泡样变样(类似病毒感染后的形态学变化),局部区域细胞密集(轻度增生)。结论金葡菌L型DNA已在感染的体内、外细胞内出现,且在诱发的肿瘤细胞内表达显著增高,推测其在诱发体细胞增生和癌变中起重要作用,很可能和病毒的致瘤机理相似。To evaluate the effect of the Staphylococcus aureus L-form infection on tumor tissues and the cultured cells, C57 mice and the monkey kidney cells MA104 strain were infected with S. aureus L-forms, and then the positive rate of L-forms found in the hepatic tissues of C57 mice and tumor tissues developed were detected by means of Gram＇ s staining, immunohistochemical and PCR assay. The patho-morphological changes in the infected monkey kidney cells were observed. It was demonstrated that the infection rate of S. aureus L-formss, expression rate of L-form antigen as well as the DNA expression rate in normal hepatic and tumor tissues were 0% （0/10） and 70% （7/10）respectively. Vacuolar degeneration with light proliferation could be observed in the in vitro cultivated monkey kidney cells infected with elementary bodies of L-forms 48 hours after infection. These results suggest that the L-forms of S. aureus may play an important role in the induction of somatic cell proliferation and development of tumor ceils.
