摘  要:目的探讨金黄色葡萄球菌(金葡菌)L型感染C57小鼠致瘤后,Mdm2癌基因及PTEN、p53抑癌基因在小鼠肿瘤中的表达及相关性研究。方法动物实验观察金葡菌L型感染90只C57小鼠后10只小鼠发生肿瘤;13只小鼠引起癌前病变。革兰染色、免疫组化及原位杂交检测小鼠肿瘤和癌前病变中金葡菌L型检出率和Mdm2、PTEN、p53蛋白及cDNA基因的表达。结果10只小鼠肿瘤及13只小鼠癌前病变中金葡菌L型及其cDNA基因阳性表达与正常对照组有明显差异(P<0.005-0.05);Mdm2、p53蛋白和cDNA基因的阳性表达与正常对照组有明显差异(P<0.05),呈正相关。PTEN蛋白和cDNA基因的阳性表达小鼠肿瘤及癌前病变与正常肝组织对照有显著性差异(P<0.01-0.05),呈负相关。结论金葡菌L型感染可能与Mdm2、PTEN、p53基因协同表达,在小鼠肿瘤发生和发展中起重要作用。To investigate the correlation of Mdm2 ,PTEN and the tumor suppressor gene p^53 expression in mouse tumor cells to infection with L-form of Staphylococcus aureus ,the occurrence of mouse tumors in C57 mice infected with L-forms of S. aureus was observed, in which 10 out of 90 mice infected developed mouse tumors, and the precancerous lesions could be demonstrated from 13 out of 90 mice infected. By using immunohistochemical assay and in situ hybridization, the expressions of Mdm2, TPEN and p^53 protein and cDNA could be detected in these lesions. In addition, the positive rate of infection with S. aureus L-form was positively correlated with the expression of Mdm2, PTEN and p^53 cDNA (P〈0. 005-0.05)and these expressions showed significant difference with that of normal controls (P〈0. 05). The positive rate of PTEN and cDNA in these cases was negatively correlate with normal control group (P〈0. 05). It is concluded that the expression Mdm2, PTEN and p^53 gene in mouse tumor cells correlate with the infection of S. aureus L-forms, suggesting that it may play a role on the development of tumors in mice.
