人胚绒毛提取液对NCI446细胞生长的影响及其作用机制

摘　　要：目的探索人胚绒毛组织提取液（Human Embryonic Chofion Extract，HECE）中是否存在抑瘤物质。方法本研究将妊娠30～50d左右的人胚绒毛组织制成匀浆，收集上清，即人胚绒毛提取液（Human Embryonic Chofion Extract，HECE），采用MTT法检测其NCI446（人小细胞肺癌）肿瘤细胞株的生长作用。本实验对NCI446细胞进行了系统的研究：HECE对NCI446细胞不同时间的生长作用，凋亡的检测及对转染人类多药耐药基因（MDR1）的NCI446细胞的生长作用。结果HECE对NCI446细胞有明显的抑制作用，呈剂量依赖关系，NCI446细胞与HECE共育48h，原液组存活率为58．10％，延长对NCI446细胞作用时间至72h，仍有抑制作用，但结果与48h基本一致。采用TUNEL法检测凋亡，可见凋亡细胞，阳性率可达31％（3次实验），对已转染MDR1基因的NCI446细胞这种抑瘤作用降低。结论这种提取液中存在着天然的抑瘤成份，这种抑制作用是通过诱导细胞凋亡实现的，但不排除可引起细胞坏死。Objective It is well known that the chorion tissue feeds the development of embryo. Are there any tumor suppressors or other cytokines in the chorion tissue？ and how to work？ Until now, there is no report about biological activity of human embryonic chofion in the references Methods Human embryonic chorion extract （HECE） was prepared in this study. The effect of HECE on the growth of NCI446 （human lung cancer cell line） was detected using the method of MTF, In the study, we have performed the systemic experiments on NCI446 cells including the effect of HECE on the growth of NCI446 cells in different time, apoptosis detection, and the effect on the growth of NCI446 cells transfected with MI）RI. Results It was demonstrated that HECE showed preferentially inhibitory effects on the growth of NCI446 cells, and which appeared dose-dependent relationship, In order to understand what the mechanism is, the apoptosis cells of NCI446 after exposed to HECE were counted using TUNEL, the positive rate was about 31% （three experiments）, the negative control were no positive cells. The result showed that this suppressive effect induced by cell apoptosis. This suppressive effect decreased on the growth of NCI446 transfected with MDR1, Conclusions The experiment data mentioned above show that the natural suppressors exist in the human embryonic chorion tissue, it has inhibitory effect on NCI446 cell line.
