摘 要:目的:探讨不同来源痤疮丙酸杆菌培养上清对单核巨噬细胞株THP-1细胞产生TNF-α、IL-12的影响,明确外伤后细菌性致死性肉芽肿分离菌株与标准株刺激机体产生细胞因子的差别。方法:采用ELISA法、RT-PCR的方法观察标准株与从外伤后细菌性致死性肉芽肿分离株的培养上清对THP-1细胞产生TNF-α和IL-12的影响。结果:(1)痤疮丙酸杆菌培养滤液可促进THP-1细胞产生TNF-α和IL-12,和空白对照组相比有显著性差异(P<0.05),标准株较临床株有较强的刺激THP-1产生TNF-α的能力;(2)所有培养上清均能促进THP-1细胞产生IL-12,标准株较外伤后细菌性致死性肉芽肿分离株培养滤液的刺激THP-1细胞产生IL-12作用弱(P<0.05)。结论:不同来源痤疮丙酸杆菌培养上清均可促进THP-1细胞产生TNF-α、IL-12,但其能力不同,说明不同来源痤疮丙酸杆菌刺激机体早期免疫应答水平不同。Objective: To investigate the influence of different Propionibacterium acnes culture supernatants on the production of TNF - α and IL- 12 by THP - 1 cells and to determine the difference in the production of cytokines induced by the P. acnes isolated from fatal bacterial granuloma after trauma (FBGT) and standard strain of P. acnes. Methods: The influence of supernatants obtained from standard strain and FBGT culture on the production of TNF- α and IL- 12 by THP- 1 was determined by ELISA and RT- PCR methods. Results: The supernatants of P. ances culture significantly enhanced the production of TNF - α and IL- 12 by THP - 1 cells when compared with controls ( P 〈 0.05). For TNF - α the enhancement of standard strain was stronger than that of FBGT, while for IL - 12 FBGT was stronger than standard strain. Conclusion : Different P. ances stains have different capacities of enhancing the production of TNF- α and IL- 12 by THP- 1 cells.
