摘 要:目的探讨在外伤性脑损伤的条件下,移植同种异体神经干细胞所诱发的淋巴细胞浸润现象。方法60只SD大鼠随机分配到实验组(n=30)和对照组(n=30),制作脑损伤模型,实验组将Wistar大鼠来源的神经干细胞移植到损伤灶,对照组注射等量生理盐水。通过脑组织切片HE染色、免疫组织化学以及流式细胞术检测局部淋巴细胞浸润的表现,并通过流式细胞术检测神经干细胞的MHC-Ⅰ分子。结果神经干细胞表达MHC-Ⅰ分子的阳性率为6.57%±0.44%。HE染色和免疫组织化学显示,实验组脑损伤灶有相当数量的淋巴细胞浸润,对照组未见淋巴细胞浸润。流式细胞术显示,在损伤脑组织中,实验组的CD4^+淋巴细胞数/总细胞数比率为13.28%±1.60%,对照组为0.41%±0.12%,P〈0.01);实验组的CD8^+淋巴细胞/总细胞数比率为5.11%±1.03%,对照组为0.57%±0.26%,P〈0.01)。结论神经干细胞具有一定免疫原性,当移植到外伤性脑损伤病灶中时,可以诱发局部淋巴细胞浸润。Objective The process of immune response to neural stem cell allograft has not been fully understood. This study was designed to investigate the lymphocyte infiltration induced by transplantation of neural stem cells into traumatically injured brain. Method Sixty SD rats were assigned randomly to the transplantation group ( n = 30) and the control ( n = 30) . All animals were subjected to focal brain contusion following the Feeney's method. Then the neural stem cells derived from newborn Wistar rats were transplanted into the traumatically injured brains. As for the rats of the control group, they were injected saline instead. Lymphocyte infiltration in the lesion was studied by means of HE staining, immunobistochemistry and flow eytometry. The immunogenieity of neural stem cells was evaluated by their expression of MHC-Ⅰ molecules, as shown by flow cytometry. Results About 6.57% ± 0.44% of neural stem cells expressed MHC-Ⅰ molecules. Histological examination and immunohistochemistry revealed that many lympbocytes infiltrated in the adjacent area of the contusion. No similar lymphocyte infiltration was found in the control group. As shown by flow cytometry, in the traumatic brain tissues of the transplantation group, the cell number percentage of CD4^+ lympbocytes was 13.28% ± 1.60%, and that in the control group was 0.41% ± 0.12%, P 〈 0.01. Likewise, the percentage of CD8^+ lymphocytes of the transplantation group was 5.11% ± 1.03%,and that of the control was 0.57% ± 0.26%, P 〈 0.01. Conclusions Neural stem cells are inherent in immunogenicity. When transplanted into traumatically injured brain, they can induce lymphocyte infiltration.
