摘  要:目的:研究人参皂甙Rg1、Rb1对体外培养大鼠SVZa-NSCs(室管膜前下区神经干细胞)增殖的影响及其与STAT3(信号转导与转录激活因子3)的关系。方法:采用体外细胞培养法对大鼠SVZa-NSCs分离、培养,用细胞计数法和MTT法测定人参皂甙Rg1、Rb1对NSCs增殖的影响,并寻找促进NSCs增殖的人参皂甙Rg1、Rb1最佳浓度和最佳时间;用免疫细胞化学法检测STAT3的表达和阳性细胞百分比,NSCs增殖与JAK/STAT通路的关系。结果:人参皂甙Rg1、Rb1能够促进SVZa-NSCs的增殖,其最佳浓度为40μM、最佳时间为4d。与对照组比较,人参皂甙Rg1、Rb1明显促进STAT3的表达和阳性细胞百分比增加(P<0.01)。结论:人参皂甙Rg1、Rb1对内源性NSCs的增殖、分化有较大价值,为中枢神经系统损伤修复提供实验基础理论。Objective: To study the effect of Ginsenoside Rgl ,Rbl on the proliferation of in vitro cultured SVZa-NSCs in rats and its relation with the expression of STAT 3. Methods: SVZa-NSCs in rats were cultured in vitro and isolated. The effects of Ginsenoside Rgl and Rbl on the proliferation of NSCs were tested by MTT and cytometry. The optimal concentration and the optimal time for Ginsenoside Rgl and Rbl enhancing the proliferation of NSCs were determined. The expression of STAT 3 and the percentage of positive cells were tested by immunocytochemistry. The relationships between the proliferation of NSCs and the JAK/STAT pathway was explored. Results: Ginsenoside Rgl and Rbl could enhance the proliferation of SVZa-NSCs. The optimal concentrations of Ginsenoside Rgl and Rbl to enhance the proliferation of NSCs tested by cytometry were both 40μm, and the results tested by MTT were 20 μM and 40 μM respectively. Ginsenoside Rgl and Rbl could enhance the expression of STAT 3 and significantly increase the percentage of positive cells(P〈0.01). Conclusion: Ginsenoside Rgl and Rbl have a significant value for the proliferation and differentiation of endogenous NSCs,and provide the experimental and theore-tical basic therapy for the rehabilitation of central nervous system injury.
