大鼠异体肢体移植中淋巴细胞亚群的动态观察

摘　　要：目的探讨大鼠后肢异体移植中排斥反应与T淋巴细胞亚群和活化T细胞的关系。方法将24只(8只Wistar,16只SD)大鼠随机分成两组,实验组8只为Wistar→SD后肢移植未作免疫抑制组,对照组8只为FK506(2mg/kg体重)和霉酚酸(20mg/kg体重)免疫抑制组,观察术后排斥反应及病理检查;流式细胞仪检测T细胞亚群和活化T细胞。结果术后6～8 d,实验组移植肢体出现排斥反应,活化T细胞(CD3/CD69、CD3/CD25、CD3/HLA-DR)已于排斥反应发生(6.88±0.83)d前2～4d显著升高,CD4/CD8比值中度升高;活化T细胞增高与排斥反应程度呈正相关(相关系数r为0.874、0.854、0.879,P<0.05);CD4/CD8比值与排斥反应程度无明显相关(相关系数r=0.544,P>0.05);对照组,活化T细胞较术前无明显变化,未出现排斥反应。结论动态监测活化T细胞将有助于异体肢体移植急性排斥反应的早期诊断。Objective To investigate the relation between the T lymphocyte subsets, activated T cells and acute rejection in the rat limb allotransplantation. Methods Twenty - four male rats were randomly divided into two groups ： experimental group （ n = 8） and control group （ n = 8 ）. In experimental group,Wistar→SD limb allotransplantation was performed without drug administration, while in control group ,the recipients were immunosuppressed with FKS06 and MMF. The rejection and histologic degree of skin and muscle rejection of the limb transplants were explored. T cell subsets and activated T cells were also determined by flow cytometry. Results In experimented group, activated T cells （ CD3/CD69, CD3/ CD25 ,CD3/HLA-DR） were increased significantly 2-4 days before the onset of rejection （6.88±0.83 days） and positively related to the degree of the rejection （correlation coefficient was 0. 874,0. 854, 0.879, P 〈 0.05 ）. Although the ratio of CD4/CD8 was increased modestly after the rejection ,there was no evidently relation between the ratio and rejection （ correlation coefficient was 0.544, P 〉 0.05 ）. In control group,there were no significant changes in the ratio of CD4/CD8 and activated T cells after operation and no rejections occurred. Conclusion The dynamic assay of activated T cells （ CD3/CI）69, CD3/CD25, CD3/HLA-DR） is helpful to the early diagnosis of rejection of rat limb allotransplantation.
