摘 要:目的:采用高效液相色谱法(HPLC)对丹参超微饮片水溶性有效部位进行指纹图谱研究。方法:采用Hypersil BDS-C18柱(4.6mm×250mm,5μm),乙腈-0.014(体积分数)磷酸溶液梯度程序洗脱,流速1.0mL/min,柱温25℃,检测波长280nm。以27个共有峰为评价指标,使用“中药色谱指纹图谱相似度评价系统2004A版”软件计算处理。结果:建立的指纹图谱测定系统,对丹参水溶性成分分离良好。将8个产地的10批药材的指纹图谱进行相似度评价,相似系数均在0.9以上。结论:该法操作简便,精密度、稳定性和重复性良好,为丹参超微饮片水溶性有效部位的质量控制提供了方法。AIM: To establish the fingerprint chromatograms for the quality evaluation of Salvia miltiorrhiza Bge. by HPLC. METHODS : The analysis was performed on a Hypersil BDS-C18 column (5 μm, 4.6 mm × 250 mm) with acetonitrile-water (containing 0. 014% (v/v) phosphat acid) as mobile phase at a flow rate of 1.0 mL/ min, and at a column temperature of 25 ℃. The detection wave-length was at 280 nm. The mutual fingerprint of the extract of Salvia miltiorrhiza Bge was established by using the similarity calculation software of Chinese herbal fingerprint. RESULTS: The HPLC fingerprints of the water-soluble extract of reference S. miltiorrhiza were obtained with very good resolution under the established chromatographic system. The quality of ten batches of S. miltiorrhiza samples from 8 habitats were assessed by comparing their chromatographic fingerprints with the reference fingerprints obtained at the same time, and the similarity showed no difference. CONCLUSION: The method is reliable and can be helpful in effectively controlling the quality of Salvia miltiorrhiza Bge.
