摘 要:目的:研究“丹参饮”中使药(檀香)对君药(丹参)药代动力学参数的影响。方法:建立家兔含药血浆中丹参素含量的高效液相色谱-电喷雾-离子阱质谱(HPLC-ESI-MS/MS)分析方法。血浆用10%高氯酸沉淀蛋白,上清液用乙酸乙酯萃取获得样品。以对羟基苯甲酸为内标,采用离子阱质谱的多级反应模式(MRM)测定丹参素的血药浓度,DAS数据处理软件对数据进行统计学处理。结果:丹参素在0.2-40.0μg/mL范围内线性关系良好,方法检测限为1.0 ng,重复性实验RSD(%)为1.34(n=6),高、中、低3个浓度的平均回收率(%)分别为(80.69±3.93),(95.5±2.81),(74.21±3.58)。配伍给药前后,丹参中丹参素体内代谢动力学模型均为二室开放模型,但配伍给药后AUC0-∞较丹参单独给药明显增大。结论:“丹参饮”中使药(檀香)可促进君药(丹参)吸收入血,该研究可为“君-使”对药体内作用机制的研究提供新思路。Aim: To study the influence of Santalum album (Shi herb) on the pharmacokinetics of Radix Salvia miltiorrhizae (Jun herb ) in the rabbits receiving Danshenyin. Methods: A valid assay for the determination of Danshensu was established by HPLC-ESI-MS/MS. The plasma was pretreated with 10% perchlorie acid and the supernatant was extracted by ethyl acetate. The p-hydroxybenzoic acid was used as an internal standard to detect the blood-drug concentrations by HPLC-ESI-MS/MS. The collected data were processed by computer program DAS to study the effects of Santalum album on the pharmaeokineties of Radix Salvia miltiorrhizae. Results: The linear range of Danshensu in the assay was 0.2 - 40.0 μg/mL. The limit of detection (LOD) was 1.0 ng. The RSD as reproducibility in experiment was 1.34% ( n = 6). The average recovery at lower, middle, and upper spiked concentrations was (74.21 ± 3.58) %, (95.5±2.81 ) % and (80.69±3.93 % ), respectively. Moreover, prior to and post-oral dosing of compatibility, the pharmacokinetics of Dansbensu was found to be fitted to open two-compartment model. And co-administration resulted in obvious increase in AUCo-∞. Conclusion: Shi herb could promote the absorption of Jun herb in Danshenyin. The results could provide clues for understanding of phamaaeologieal mechanism of Jun-Shi pair herbs.
