摘 要:目的人类白细胞抗原(Human Leucocyte Antigen,HLA)编码基因具有高度遗传多态性,有关中国人群HLA分布,已有的文献报道主要为汉族人群数据。本工作首次采用DNA分型技术,分析蒙古族HLA-A位点多态性。方法采用ARMS/PCR技术对83例居住在内蒙古地区的蒙古族健康个体做HLA-A DNA分型。结果该人群存在HLA-A*01至A*33等11种等位基因(组),其中,以HLA-A*02频率最高(0.2886),HLA-A*11、A*24次之;该人群中,HLA-A19宽特异性(Broad Specificity)含有HLA-A*29、A*30、A*31、A*32、A*33等5组编码基因,频率之和为0.2368;此外,在蒙古族,HLA-A*01较为常见(基因频率为0.08823)。结论本研究在DNA水平提供了蒙古族HLA-A位点频率分布数据。Objective The genes encoding human leucocyte antigen are highly polymorphic, however, previous studies regarding the HLA distribution in Chinese populations have mainly focused on Chinese Han populations residing at various geographic regions. Our study was designed to explore the HLA- A genetic polymorphism of Chinese Mongolian population at DNA level. Methods Eighty - three unrelated healthy individuals of Mongol, recruited from Inner Mongolia, were typed for HLA- A locus by using amplification refractory mutation system - polymerase chain reaction(ARMS/PCR) technique. Results Eleven HLA- A alleles(groups) were found, the most frequent alleles were HLA - A * 02, A * 11, and A * 24, with gene frequencies of 0. 2886, 0. 1713, and 0. 1569 respectively. In this sample panel, HLA- A19 broad specificity was composed of HLA- A* 29, A * 30, A * 31, A * 32, and A* 33 splits, which were summed to a frequency of 0. 2368. In addition, HLA - A * 01 was a relatively common allele with gene frequency of 0. 08823. Conclusion Our study provides the data of HLA- A genetic polymorphism of a Chinese Mongolian population at DNA level.
