摘 要:[目的]探讨甲醛所致急性肺水肿动物模型的建立方法,为甲醛急性中毒肺水肿的治疗及机制研究提供实验动物模型。[方法]大鼠共20只随机分为对照组和甲醛组,称重后放入小铁笼并置于自制染毒缸,缸内放置钠石灰50g,缸内对角分别放置2个盛有50ml40%甲醛原液的培养皿,并通过接有导管的加氧泵持续给甲醛原液内通入氧气,为大鼠供氧并促进甲醛挥发。大鼠在缸内连续吸入甲醛蒸汽染毒2h,待5h后处死,取心、肺称重并计算心肺系数、肺体系数,肺组织作常规病理检查。对照组用生理盐水,处理方法同甲醛组。[结果]与对照组比较,甲醛组心肺系数显著增大,分别为1.747±0.162和1.577±0.095,P<0.05,肺体指数也显著增大,分别为5.38±0.60和4.67±0.34,P<0.01。病理切片显示肺泡内大量液体及肺泡间隔增宽等病理改变。[结论]甲醛挥发吸入能成功地复制大鼠急性肺水肿动物模型。[ Objective ] To explore the method of establishing an animal model of acute pulmonary edema induced by formaldehyde inhalation. [ Methods ] Rats were divided into normal saline control group (n=10)and formaldehyde inhalation group( n=10 )randomly, and then put in small iron cages which were placed in a self-made intoxication chamber, with preset 50 g dry sodalime and two 50 ml euhure plates filled with 40% formaldehyde liquid. Oxygen was supplied consecutively via a rubber pipe with one end connected to air pump and another end put under the formaldehyde liquid to promote the formaldehyde evaporating. The rats inhaled formaldehyde air consecutively for 2 hours and were killed 5 hours later. Lungs and hearts of rats were cut and weighed, and the eardiopulmonary index and lung index were calculated, the lung tissue were sent for routine pathologic examination. Rats of control group were treated the same way as formaldehyde inhalation group, but used normal saline instead of formaldehyde liquid. [ Results ] Compare with normal saline group, cardiopulmonary index( 1.747 ± 0.162 vs 1.577 ± 0.095, P 〈 0.05 )and lung index( 5.38 ± 0.60 vs 4.67 ± 0.34, P 〈 0.01 )increased significantly in the formaldehyde group. The pathological section of.the lungs showed that pulmonary alveoli fill with some fluid and their septum widened in the formaldehyde group rats. [ Conclusion ] Formaldehyde vapour inhalation can induced acute pulmonary edema rat model successfully.
