摘 要:目的探讨补骨脂素(PSO)加长波紫外线(UVA)光化学疗法对人白血病细胞株NB4、HL-60、K562的作用。方法以NB4、HL-60、K562细胞株为研究对象,以细胞抑制率、凋亡率及在电镜下细胞超微结构改变为检测指标,观察PSO加波长360nm的UVA对人白血病细胞株的影响,并分析各因素间的交互作用。结果当PSO浓度均为80μg/ml,UVA照射时间分别为10min、15min和15min时,NB4、HL-60、和K562细胞的抑制率分别达峰值;当PSO浓度分别为40μg/ml、80μg/ml和80μg/ml,UVA照射时间为10min、15min和15min时,NB4、HL-60和K562细胞的凋亡率达峰值。不同PSO浓度、不同UVA照射时间对不同白血病细胞株的抑制率、凋亡率的差异有统计学意义,且各因素之间有交互作用。电镜下观察经PSO和UVA处理后的NB4、HL-60及K562细胞超微结构,均可见明显的凋亡形态学改变。结论PSO加UVA(PUVA)可抑制白血病细胞株NB4、HL-60、K562的生长,并可诱导其凋亡。浓度为40~80μg/mlPSO与波长为360nm的UVA照射10~15min是PUVA抗NB4、HL-60、K562白血病细胞的最佳作用点。Objective: To explore the effects of psoralen and long wave uhraviloet A (PUVA) on NB4, HL - 60 and K562 cells. Methods: The NB4, HL - 60 and K562 cells were taken as the study objects, and the inhibition ratios, the apoptosis ratios and the ultrasturcture change were detected in order to observe the effects of psoralen ex- tracted from chinese medicine and ultraviolet A (UVA) of wave length 360 nm on human leukemia cells. The factorial design and analysis of variance were used to analyze the interaction among the factors. Results: When the concentration of psoralen was 80ug/ml and the exposure time of UVA was 15, 15 and 10 minutes, the inhibition ratios of NB4, HL - 60 and K562 cells reached the peak respectively. When the concentration of psoralen was 40, 80 and 80ug/ml and the exposure time of UVA was 10, 15 and 15 minutes, the apoptosis ratios of NB4, HL - 60 and K562 cells reached the peak respectively. There were statistically differences of cells' inhibition ratios and apoptosis ratios among the PSO with different concentration and UVA with different exposure time. And there were interaction among the factosr. After NB4, HL - 60 and K562 cells had been treated with PUVA, they all showed obvious ultrastructure change about apoptosis observed under the electron microscope. Conclusion: PUVA can inhibit the growth of NB4, HL - 60 and K562 cell lines and induce their apoptosis. The point that the concentration of PSO is 40 - 80 μg/ml and the exposure time of UVA is 10 - 15 min can be considered as the most effective point of PUVA on NB4, HL - 60 and K562 cell lines.
