摘 要:目的:建立测定血浆中氨氯地平血药浓度的HPLC—MS法,为临床用药提供参考。方法:血浆中样品用乙酸乙酯提取,HypersilODS柱(4.6mm×150mm,5μm)为分析柱,甲醇-(10mM)醋酸铵水溶液(84:16,v/v)为流动相,氨氯地平的检测离子为[M+Na]^+,m/z431.4;内标哌罗匹隆检测离子为[M+H]^+,m/z427.5。结果:血浆中氨氯地平的线性范围为0.18~18.2μg·L^-1,批内、批间误差RSD均小于10%,绝对回收率在89.5%-95.2%。结论:本法测定血浆中氨氯地平血药浓度准确、简便,适用于氨氯地平药代动力学的研究。Objective:To establish an analytical method for the determination of amlodipine in human plasma with HPLC -MS, which might provide useful information for clinical practice. Methods: Amlodipine was extracted with ethyl acetate. A Hypersil ODS column was used with mobile phase consisting of methanol-water (containing 10 mmol· L^-1 ammonium acetate) (84: 16, v/v). The ions for SIR analysis of amlodipine and internal standard perospirone ions were selected at m/z 431.4 and m/z 427.5, respectively. Results .The calibration curve was linear within the range of 0. 18 - 18.2 μg·L^-1. The RSDs of inter-day and intra-day were less than 10% and the recovery was between 89.5% and 95.2%. Conclusions: The method is simple with high sensitivity and good precision and has proved to be suitable for the pharmacokinetic study of amlodipine.
