摘  要:目的 探讨半乳凝素-3(Galectin-3)在人类子宫内膜容受性形成中的作用.方法 采用人子宫内膜上皮细胞株RL95-2进行体外培养,应用RNA干扰技术(siRNA)抑制Galectin-3表达,RT-PCR法和western blot测定Galectin-3 mRNA和蛋白表达水平.体外观察细胞黏附率的变化,并用流式细胞术观察Galectin-3表达抑制对细胞生长和整联蛋白β1表达的影响.结果 Galectin-3 siRNA转染效率达到70%~90%.siRNA转染后Galectin-3 mRNA和蛋白表达水平可下调70%~90%.Galectin-3表达抑制后RL95-2细胞G1期上升,S期下降;整联蛋白β1表达降低;对细胞外纤维黏连蛋白(Fibronectin,FN)黏附性显著降低.结论 Galectin-3可以影响子宫内膜上皮细胞的增殖、黏附,对黏附性的影响可能与调节整联蛋白的表达有关.Galectin-3可能通过调节细胞增殖、黏附等过程参与子宫内膜容受性的形成.Purpose To investigate the function of Galectin-3 in the establishment of human uterine receptivity. Methods Human endometrial cell line RL95-2 was used. The role of Galectin-3 in RL95-2 cell monolayer was examined by RNA interference technology using Galectin-3 specific small RNA. Expression of Galectin-3 was detected by reverse transcriptase-polymerase chain reaction and Western blot analysis. Cell cycle changes and the expression of integrin β1 were detected by flow cytometry. The adhesive ability of RL95-2 cells was analyzed by adhesion test. Results Galectin-3 siRNA transfection efficiency reached 70% -90%. Expression of Galectin-3 mRNA and protein in RL95-2 cells was strongly inhibited by 70% - 90% after RNA interference. Inhibition of Galectin-3 expression decreased S-phase but increased G1 phase cells. Integrin β1 expression was down-regulated, and the adhesive ability of RL95-2 cells to fibronectin (FN) was significantly reduced. Conclusions Galectin-3 is possibly involved in the establishment of uterine receptivity by regulating proliferation and adhesion of endometrial cells. The influence on adhesion may be related with the integrin modulation.
