摘 要:目的:研究丹参对移植脾再灌注损伤的保护作用及其机制。方法:采用套管法建立大鼠脾移植模型。分别用丹参加乳酸林格液、WMO-1号液、丹参加WMO-1号液灌注保存移植脾:保存2h后重建移植脾血供:再灌注3 h后切取移植脾。比较三组移植脾的阳性凋亡细胞数、细胞间黏附分子-1(ICAM-1,CD54)免疫阳性微血管数、超氧化物歧化酶(SOD)活力值和丙二酰二醛(MDA)的含量。结果:与其他两组相比,丹参加WMO-1号液灌注保存组移植脾的阳性凋亡细胞数、ICAM-1免疫阳性微血管数、MDA含量显著降低(P<0.01),SOD活力值显著增高(P<0.01)。结论:丹参通过抗自由基、抑制细胞凋亡、降低ICAM-1表达等机制对移植脾再灌注损伤起保护作用。Objective To evaluate the protective effects of radix salviae miltiorrhizae(RSM) on the ischemia-reperfusion injury of spleen graft in the rat, and to explore its possible mechanism. Methods The rat model of spleen transplantation was established, and the spleen grafts were perfused and preserved in ①RSM+lactated Ringer (LR) solution, ② WMO-1 solution, or ③RSM+WMO-1 solution respectively for 2 hours; then splenectomy was performed after 3 hours of reperfusion. Immunohistochemical staining and biochemical assays were used to evaluate the status of apoptosis, intercellular adhesion molecule (ICAM)-1 expression, activity and amount of superoxide dismutase (SOD) and malondialdehyde (MDA). Results Apoptosis, ICAM-1 expression and MDA concentration were significantly decreased in the RSM+WMO-1 solution group than the other groups(P〈0.01). The activity of SOD was significantly increased in the RSM+WMO-1 solution group(P〈0.01). Conclusions RSM+WMO-1 solution demonstrates stronger protective effects on the ischemia-reperfusion injury in the rat model of spleen transplantation. Its possible mechanism might be related to free radical antagonization, apoptosis inhibition and ICAM-1 over-expression.
