雌激素促进小儿血管瘤血管生成的体外实验研究

摘　　要：目的 论证雌激素促进dgh血管瘤血管生成的作用。方法 应用已建立的三维血管瘤血管生成体外培养模型，以雌激素及雌激素受体竞争性抑制剂三苯氧胺分组干预小儿血管瘤体外血管生成过程。实验分为四组：①组1对照组；②组2雌激素组（50pg/ml）；③组3三苯氧胺组（10^-6mol/L）；④组4雌激素（50pg/ml）＋三苯氧胺组（10～mol/L）。比较各时间点各组间新生血管区面积的差异。结果 三维血管生成模型中对照组培养2—3d后组织块芽生出细小血管，至第8—9天长成树枝状血管。培养后第3、6,9天组织块新生血管区面积组2均大组1，组4及组3均小于组2及组1（P〈0．05）。结论 雌激素能促进小儿血管瘤血管生成，从而促进血管瘤增殖，该过程能被三苯氧胺所阻滞。Objective To demonstrate that estrogen stimulates the angiogenesis of children＇ s bemangioma. Methods A piece of bemangioma biopsy was embedded in fibrin gel, and a model in vitro of angiogenesis of human bemangioma was then established. The angiogenesis of hemangioma in each group was interfered by the estrogen and tomaxifen. There were four groups divided into the followings： the group with estrogen, the group with tomaxifen, the group with estrogen ＋ tomaxlfen and the control. The dimension of newborn tubule area in the 3rd, 6th, 9th day after the culture was calculated to compare statistically differences among the groups. Results In the model of angiogenesls of bemangioma, mlcrovessels grew out from the tissue sample in 2 to 3 days after the culture, and in 8 to 9 days a complex network of microvessels had been shown, the tending to inactivity. On the 3rd,6th and 9th day after the culture the dimension of newborn tubule area of the group of estrogen[ （2.84 ± 0.20） mm2 , （ 12.93 ±0. 85 ） mm2 , （22.47± 1.40） mm2 ] were larger than those of the control[ （ 1.98 ±0. 17 ） mm^2, （7.51 ±0. 48） mm^2 , （ 11.26±0. 73 ） mm^2 ]. Those of the group of estrogen ＋ tomaxifen[ （ 1.08 ±0. 11 ）mm^2 , （3.54 ±0. 31 ） mm^2 , （5.72 ± 0.40mm^2 ] and the group of tomaxifen [ （ 1.13±0. 14） mm^2 ,（4.26± 0.29） mm^2 ,（6.08 ± 0.42） mm^2 ]were smaller than those of the groups of the estrogen and the control（ P 〈 0.05）. Conclusions The estrogen may stimulate the angiogenesis of children＇s bemangioma, and the tomaxifen may reverse the process.
