基于MCP-1的不同干预方式对病毒性心肌炎的影响作用

摘  要:目的:比较N端2-8氨基酸缺失的突变型MCP-1(mMCP-1)干预和抗-MCP-1抗体干预对病毒性心肌炎小鼠发病的影响作用。方法:CVB3腹腔注射BALB/c小鼠,随机分组处理:病毒性心肌炎模型组、抗体干预组、羊IgG对照组、mMCP-1干预组和空质粒对照组,并设未感染生理盐水对照组。采用心肌注射的方式在心肌转移抗体或mMCP-1真核表达质粒pVMm。观察小鼠心脏重量和体重的比值(HW/BW)、血清CK-MB水平、心肌组织病理学积分(PS)、心肌组织病毒载量和心肌浸润的单个核细胞变化。结果:经20μg、40μg和60μg pVMm干预的小鼠,心肌组织病变和心肌损伤较模型组减轻;不同pVMm干预剂量对心肌组织浸润的单个核细胞数的影响作用不同,40μg或60μg pVMm干预的小鼠,心肌组织中浸润的单个核细胞数明显低于模型组,20μg pVMm干预的小鼠,心肌组织中浸润的单个核细胞数与模型组相比没有明显的差异。抗-MCP-1抗体干预和mMCP-1干预小鼠相比,HW/BW、血清CK-MB水平和PS未见明显差异。结论:mMCP-1和抗-MCP-1抗体均可缓解病毒性心肌炎心肌组织病变和心肌损伤,mMCP-1可替代抗-MCP-1抗体用于病毒性心肌炎的干预治疗。Objective: To investigate the intervention of monocyte chemoattractant protein-1 ( MCP- 1 ) deleted the 2-8 amino acid in N-terminus( mutated MCP-1 ,mMCP-1 ) and anti-MCP-1 antibody with the pathogenensis of viral myocarditis induced by coxsackievirus group B type 3 (CVB3). Methods: BALB/c mice inoculated intraperitoneally with CVB3 were divided into different groups including mice model group with viral myocaditis infected alone with CVB3 , intervention group with anti-MCP-1, control group with goat IgG, with plasma pVAX,meantime the normal control designed intervention group with mMCP-1, control group group inoculated intraperitoneally with saline was . Antibody and pVMm plasmids carrying the gene of mMCP-1 were injected into myocardial tissue. Ratio of murine heart weight to body weight (HW/BW), myocardial tissue pathological score (PS) of cardiac tissue, the level of creatine kinase-MB fraction (CK- MB) in serum and CVB3 loading of myocardial tissue were analyzed. Results : Compared with the model group mice, the cardiac lesion and injury of the mice were alleviated when interfered with 20 p,g,40 p,g or 60 μg pVMm. Different dose of plasmid pVMm showed different effect on the infiltration of the mononuclear cells into myocardial tissue. Compared with model group mice, the number of the infihraflag mononuclear cells in myocardium with viral myocarditis was decreased when transferred myocardially with 40 μg or 60 μg pVMm. While there was no difference between the model group mice and the mice interfered with 20 μg pVMm in the infiltration of mononuclear cells. And there was also no difference between the mice interfered with anti-MCP-1 antibody and mMCP-1 in ratio of HW/BW, the level of CK - MB in serum and PS. Conclusion : Both mMCP-1 and anti-MCP-1 antibody could alleviate the cardiac lesion and cardiac injury of mice with viral myocarditis, mMCP- 1 could be used for the therapy of viral myocarditis instead of anti-MCP-1 antibody.
