产甘油假丝酵母基因CgZWF片段的克隆与序列分析

摘　　要：以产甘油假丝酵母基因组DNA为模板,通过简并PCR和反向PCR扩增得到了一个长为1 244 bp片段,并将其克隆到pMD18-Tvector。基因序列分析表明,此片段编码的氨基酸序列与Kluyveromyces lactis的G6pdp同源性最高(62.7%),表明已经得到编码葡萄糖6-磷酸脱氢酶的目的基因片段。该研究为阐明磷酸戊糖途径在产甘油假丝酵母耐高渗和甘油高产中的作用奠定了基础。A fragment of 1244 bp was obtained from Candiad glycerinogenes by degenerate PCR and inverse PCR. And the fragment was cloned into pMD18-T vector. Sequence analysis showed that the deduced amino acid sequence of the fragment was the most homologous to G6pdp of Kluyveromyces lactis （62.7 %）. And it was confirmed that the partial CgZWF encoding glucose 6-phosphate dehydrogenase was cloned. This study facilitated the research on the roles of pentose phosphate pathway in the osmoadaptation and glycerol metabolism in C. glycerinogenes.
