摘 要:目的观察188Re血管内照射对抑制新西兰白兔血管损伤术后平滑肌细胞增殖疗效。方法40只新西兰白兔随机分为对照组(n=20)和照射组(n=20)。照射组行腹主动脉球囊内皮拉伤术后予188Re血管内照射,管腔下0.5mm处累计照射剂量达到15Gy;对照组行腹主动脉球囊内皮拉伤术后不予血管内照射。分别于术后3、6周处死动物,取病理组织学标本进行增殖细胞核抗原(PC-NA)和凋亡细胞(TUNEL)分析,动物照射前、后检测血常规。结果与对照组相比,照射组3周PCNA阳性细胞百分比明显下降(P=0.046),平滑肌细胞凋亡百分比明显增高(P=0.018),6周两组PCNA阳性细胞百分比、平滑肌细胞凋亡百分比相比差异无统计学意义(P>0.05);照射前、后血常规检测红细胞、血红蛋白、白细胞、血小板差异无统计学意义(P>0.05)。结论血管内照射能有效抑制新西兰白兔血管损伤术后平滑肌细胞增殖,促进平滑肌细胞凋亡,研究采用的血管内照射剂量未发现有骨髓抑制作用。Objective To observe the effect of ^188 Re intravascular irradiation on inhibiting proliferation of rabbit vessel smooth muscle cells after balloon angioplasty. Methods Forty New Zealand white rabbits were divided into two groups randomly : the control group ( n = 20) and the irradiation group ( n = 20 ). In both groups, balloon catheter overstretched on abdominal aorta artery to denude endothelium, in irradiation group alone, ^188Re-radioactive liquid filled balloon irradiation was performed in target segment immediately after balloon overstretched injury until the accumulative radioactive dose under the irradiation surface 0.5 mm was add up to 15 Gy. Target segments were harvested at 3,6 weeks after the operation respectively. Pathohistological samples were analyzed. Blood routine were detected before and after ^188Re irradiation treatment. Results In 3 weeks, as compared with the control group, PCNA positive cells, TUNEL positive cells were decreased significantly in irradiation group (P = 0. 046, P = 0. 018, respectively), but there were no significant difference between two groups in 6 weeks( P 〉 0. 05 ). There were no significant difference in the level of red blood cell, hemoglobin, white blood cell , blood platelet value before and after irradiation treatment( P 〉 0.05 ). Conclusion ^188 Re intravascular irradiation can efficiently inhibit the proliferation and accelerate the apoptosis of VSMCs after balloon angioplasty without bone marrow depression effect.
