摘　　要：目的:检测Nogo-A蛋白在纯化后的大鼠脊髓和小脑细胞核中的表达。方法:通过差速离心法得到大鼠脊髓和小脑样本的各亚细胞成分,利用非离子性去污剂NP-40纯化细胞核组分,采用Western blot方法检验核纯度并检测Nogo-A在各亚细胞成分中的表达。结果:纯化的细胞核抽提物中检测到特异的Nogo-A反应条带。结论:Nogo-A在纯化的细胞核中仍有明确表达,进一步证明了Nogo-A蛋白在中枢神经元的细胞核中有定位,并提示Nogo-A很可能以全长形式入核。Objective：To investigate the expression of Nogo-A in relatively purified nucleus fractions from rat spinal cord and cerebellum. Methods：Subcellular fractions from rat spinal cord and cerebellum were got by differential centrifugation. Non-ionic detergent NP-40 was applied to purify crude nuclear pellets. Then Western Blot was used to check the purity of nuclear fractions and the expression of Nogo-A. Results：The extracts of purified nuclear fractions demonstrated a specific Nogo-A band. Conclusion：Nogo-A does exist in purified nucleus, which further proves the intranuclear localization of Nogo-A protein and suggests that Nogo-A probably may go into the nucleus in the form of full length .
