摘 要:目的观察丹参注射液(Salviae Miltiorrhizae injections SMI)对阿霉素(Adriamycin,ADR)诱导大鼠心肌细胞损伤的保护作用并探讨其作用机制。方法将培养72h以后的新生Wister乳鼠心肌细胞随机分为正常对照组、ADR损伤组和SMI小低、中、高剂量保护组,继续培养24h后,测定培养基中乳酸脱氢酶(LDH)释放量,超氧化物歧化酶(SOD)活性,丙二醛(MDA)含量及心肌细胞超微结构。结果ADR(终浓度为1mg/L)可致培养基中LDH释放量增加(P<0.01),MDA含量升高(P<0.05)而SOD活性下降(P<0.01)。SMI中剂量(50umol/L)即可降低培养基中LDH释放量(P<0.01),降低MDA含量(P<0.01)而增加SOD活力(P<0.01),心肌细胞超微结构基本恢复正常。结论SM能够保护ADR引起的心肌细胞损伤,其机制可能与其减少氧自由基生成有关。Objective To observe the protection effect and its mechanisms of Salviae Miltiorrhizae Injection(SMT) on myocardium cell injury induced by adriamycin(ADR). Methods The neonatal Wistar rats cardiomyocytes cultured for 72 hours were divided into three groups: normal control group, ADR damnified groups and groups of Salviae Miltiorrhizae Injection for little, middle and higher doses. After 24 hours' further cultivation,the release content of cell culture medium lactate dehydrogenase(LDH), malondialdehyde(MDA) content, the activity of superoxide dismutase( SOD) and ultrastructure of myocardium cell were determined. Results ADR increased the release content of LDH (P 〈 0. 01), increased the content of MDA (P〈 0.05) and the activity of SOD (P〈 0.01)in the culture medium. The middle quantity of SMI(50umol/L) decreased the release content of I.DH (P 〈 0.01), decreased the content of MDA (P 〈 0.05) and increased activity of SOD (P 〈 0.01)in the culturing medium,the ultrastructure of myocardium recovered to the normal level generally. Conclusions SMI can protect the Myocardium Cell from injury induced by ADR,the mechanism has relationship with the reduction of oxygen free radical.
