摘  要:目的:研究周围神经损伤后神经生长因子(NGF)、睫状神经营养因子(CNTF)对神经元的保护作用。方法:以硅管套接大鼠坐骨神经,局部及皮下分别应用SAL(生理盐水)、NGF和CNTF。术后2周,应用酶组织化学方法显示相应节段脊髓前角运动神经元和脊神经节感觉神经元中胆碱脂酶(CHE)、酸性磷酸酶(ACP)酶活性并结合计算机图像分析。结果:与正常侧相比,SAL组两类神经元中CHE活性显著减弱,ACP活性显著增强;NGF组感觉神经元两酶活性变化不显著,运动神经元两酶活性有显著变化;CNTF组的结果与NGF组相反。结论:周围神经损伤后,外源性NGF主要保护感觉神经元,CNTF主要保护运动神经元。Objective:Protective effect of exogenous NGF and CNTF on neurons was studied after peripheral nerve injury.Methods:After silicone tubulization of sciatic nerve in rat, 0 9% saline,NGF or CNTF solution was injected into silicone chamber during nerve repair and subcutis every day after surgery.Enzyme histochemistry was used to show CHE,ACP activity of spinal motoneurons in spinal segments and sensory neurons in spinal ganglions combined with computer image analysis.Results:Compared with the control side,both enzymes activity significantly changed in both neurons in SAL group.Both enzymes activity significantly changed in motoneurons,but not in sensory neurons in NGF group.The results in CNTF group were contrary to that in NGF group.Conclusions:These results suggest that exogenous NGF protects sensory neurons and exogenous CNTF,motoneurons after peripheral nerve injury.
