摘 要:目的探讨对氯苯酚与DNA的相互作用。方法在0.05mol/LNa2HPO4-0.05mol/LNaH2PO4-0.10mol/LNaCl缓冲体系中(pH=7.30),研究对氯苯酚在核苷三磷酸修饰电极及DNA修饰电极上的电化学行为。结果双链DNA(dsDNA)可对氯苯酚的氧化峰正移0.032V,峰电流减小;但单链DNA(ssDNA)对对氯苯酚的峰电位改变较小,峰电流增大;对氯苯酚在三磷酸鸟苷(dGTP)、三磷酸胸苷(dTTP)和三磷酸胞苷(dCTP)修饰电极的氧化过程中于0.78V左右产生了新的氧化峰。结论dsDNA与对氯苯酚形成的∏∏共轭体系,但ssDNA的这种作用力较小;对氯苯酚在dGTP、dTTP和dCTP修饰电极的氧化过程中产生了新的加合物。Objective To study the interaction between p-ehlorophenol and DNA. Methods The electrochemical behaviors of p-ehlorophenol were determined at nuelcoside triphosphate, single chain DNA (ssDNA) and double chain DNA (dsDNA) modified electrode in 0. 05 mol/L Na2HPO4 -0. 05 mol/L NaH2PO4 -0. 10 mol/L NaCl buffer solution (pH =7. 30). Results dsDNA decreased the current of p-chlorophenol and shifted its potential to positive by 0. 032 V; however, ssDNA could increase the current of p-chlorophenol and shift slightly its potential to positive. When p-chlorophenol was being oxidized at dGTP dTTP and dCTP modified dectrode, new peaks were found near 0. 78 V. Conclusion The interaction between p-chlorophenol and dsDNA ascribes to П Пconjugate, however, this force between ssDNA and p-chlorophenol is smaller than that between p- chlorophenol and dsDNA. When p-ehlorophenol is being oxidized at dGTP,dTTP and dCTP modified electrode, new adduets are produced.
