摘  要:目的:探讨急性肺损伤大鼠肺Ⅰ型和Ⅲ型胶原合成的变化。方法:SD大鼠24只,随机分为正常对照组、潮气量(VT)8ml/kg组(A组)和16ml/kg组(B组),每组8只。A、B两组分别机械通气4h,通气结束后测定肺组织湿/干重(W/D)、肺泡灌洗液与血浆蛋白比值、Ⅰ型和Ⅲ型前胶原mRNA表达及其比值、Ⅰ型和Ⅲ型前胶原肽(PⅠNP和PⅢNP)浓度。结果:①与正常对照组肺组织W/D比较,A组和B组均显著增加(P均<0.05),B组又明显高于A组(P<0.05);②正常对照组和A组肺泡灌洗液与血浆蛋白比值无显著差异,而B组显著高于前两组(P<0.05);③Ⅰ型前胶原mRNA表达在正常对照组与A组之间无显著差异,而B组显著高于前两组(P均<0.05);④Ⅲ型前胶原mRNA表达在正常对照组与A组之间无显著差异,B组显著高于前两组(P均<0.05);⑤Ⅰ型前胶原与Ⅲ型前胶原mRNA比值3组间比较均无显著差异(P>0.05);⑥PⅠNP、PⅢNP表达3组间比较均无显著差异(P>0.05)。结论:高潮气量通气导致正常大鼠出现肺损伤,Ⅰ型和Ⅲ型前胶原mRNA表达增加,但4h内并未出现Ⅰ型和Ⅲ型前胶原比值增加,胶原蛋白尚未增加。Objective: To investigate the changes of collagen synthesis of lung type Ⅰ and type Ⅲ in rats with acute lung injury. Methods:Twenty-four male SD rats were randomized divided into three groups (n = 8) : control group, group A and group B. Group A and B were treated with different tidal volume (Vt 8 ml/kg, Vt 16 ml/kg), both received mechanical ventilation for 4 hours. Wet/dry weight ratio (W/D), ratio of protein concerntration in bronchoalveolar lavage fluid and plasma, the expressions of type Ⅰand type Ⅲ procollagen mRNA, concemtration of type Ⅲ and type Ⅲ procollagen were detected. Results. (1)W/D in group A and group B were significantly higher than that in control group(P〈0. 05), while group B was higher than that of group A(P〈0. 05). (2)There was no significant difference in bronehoalveolar lavage fluid and plasma protein ratio in control group and group A,while the bronchoalveolar lavage fluid and plasma protein ratio in group B was significant higher than that of the above two groups(P〈0. 05). (3)There was no significant difference in the expression of type Ⅰ procollagen mRNA between the control group and group A(P〈0. 05) ,while the expression of type I precollagen mRNA in group B was significandy higher than that of the above two groups(P〈0.05). (4)There was no significant difference in the expressions of type Ⅲ procollagen mRNA between the control group and group A(P〈0. 05), while the expression of type Ⅲ procollagen mRNA in group B was significantly higher than that of the above two groups (P〈0. 05). (5)There was no significant difference among all three groups in ratio between type I collagen and type Ⅲ collagen mRNA (P〉0. 05). (6)There was no significant difference among all three groups in the expression of procollagen type Ⅰ (P NP) and type Ⅲ (PⅢ NP)(P〉0.05). Conclusions: High tidal volume can induce acute lung injury and up-regulate the expressions of procollagen type
