摘  要:目的 观察17β-雌二醇、结合雌激素和雌激素受体调节剂雷诺昔芬对去卵巢大鼠恐惧记忆的影响。方法 雌性大鼠双侧卵巢切除1月后给予药物治疗,第2月进行恐惧记忆和伤害反应测定,恐惧记忆测定采用高频声音刺激308,在最后28给予电刺激,重复同样刺激,1周后测定环境和条件性线索诱导的僵直不动;伤害反应采用逐步提高电刺激电流强度,观察动物退缩、惊叫和攻击性行为的电流阈值。结果 双侧卵巢切除动物环境诱导僵直不动的时间[(76.8±9.1)8]较假手术组[(107.3±4.8)8]明显缩短(P〈0.05),而17β-雌二醇或雷诺昔芬治疗后,僵直不动的时间[(113.6±3.9)S,112±6.4)s]较手术组明显增加(均P〈0.05);结合雌激素处理后恐惧性记忆时间稍增加,但没有统计学差异。雷诺昔芬处理后退缩、嘶叫和跳跃反应的电流阈值分别为(500±44.7)μA,(816±70.3)μA和(1133±98.9)μA,较手术对照组的(183±21)μA,(336.7±21.7)μA,(483±30.7)μA明显提高(P=0.02),17β-雌二醇和结合雌激素处理对伤害性刺激的反应没有影响。结论 结果提示17β-雌二醇在促进情感性学习记忆中发挥重要作用。Objective To investigate the effects of the estradlol, estrogen or raloxifene on the fear freezing emotion learning in the ovariectomized rats. Methods The drug maintenance was taken at one month after ovariectomized surgery. Freezing was tested by a discrete stimulus and context associated with the repeated tone- footshock pairings. The electric shock thresholds (mA) to elicit stereotypic responses (flinch, vocalization, jump and vocalization) were measured. Results The ovariectomized rats produced significantly less freezing induced by fear context than that of control rats. The chronic administration with either estradiol or raloxifene for one month exhibited the enhanced acquisition of tone and context fear, but estrogen treatment did not change the fearing memory. Meanwhile, the treatment with raloxifene showed stereotypical progressive flinch, jump, and vocalization responses, and estradiol or estrogen failed to affect shock reactivity for any response. Conclusion These data suggest that estradiolor, raloxifene rather than estrogen may play an important for emotional memory during deprive of sex hormone.
