摘  要:目的:观察败血症休克大鼠血浆和肺组织中intermedin(IMD)含量和mRNA表达变化情况。方法:盲肠结扎穿孔法制备大鼠腹膜炎早、晚期败血症休克(Early sepsis,ES;Late sepsis,LS)模型,放射免疫法测定血浆和肺组织IMD含量。半定量RT-PCR测定肺组织IMD mRNA水平。结果:与对照组相比,ES大鼠血浆和肺组织IMD含量分别增加609.3%和39.9%(P<0.001),与ES大鼠相比较,LS大鼠血浆IMD浓度增高60.6%(P<0.001),而肺组织IMD浓度无明显变化(P>0.05);与对照组相比,LS大鼠肺组织IMDmRNA表达水平增高135.4%(P<0.001);但ES大鼠则无明显变化(P>0.05)。结论:IMD在败血症休克大鼠血浆和肺组织中显著增加,参与了休克的病理生理过程,但机制不明。Objective To investigate the changes of intermedin (IMD) content and mRNA level in lungs of rats with peritonitis septic shock. Methods The peritonitis septic shock model was established by cecal ligation and perforation in SD rats. Ten hours after operation was considered as early sepsis ( ES group) ,and 20 hrs as late sepsis ( LS group). Contents of IMD were determined by radioimmunoassay; the IMD mRNA was measured by semi - quantitative RT - PCR. Results Compared with control group,in the ES rats, the contents of IMD in plasma and lung homogenates were increased by 609.3% and 39.9% ( P 〈 0.001 ), respectively. Moreover, the contents of IMD in plasma were increased by 60.6% ( P 〈 0.001 ) in the LS rats than that in ES rats, but the contents of IMD in lung homogenates had no obvious change ( P 〉 0.05 ). Compared with control group, the levels of IMD mRNA in lungs in the LS rats were increased by 135.4% ( P 〈 0.001 ), but had no obvious change in the ES rats( P 〉 0.05 ). Conclusion The IMD content in plasma and lung is significantly increased in rats with septic shock, which suggests that LMD is involved in the pathogenesis of shock.
